Objective To investigate the changes of the frequency and function of CXCR5+CD8+T cell subsets in peripheral blood of HIV infected patients and their correlation with disease progression. Methods Forty HIV infected patients and 15 health controls were enrolled in this study. The frequency of CXCR5+CD8+T cells and the levels of IFN-γand IL-10 expression in peripheral blood were examined by flow cytometry. In addition, the correlation of the frequency and function of CXCR5+CD8+T cells with peripheral CD4+T lymphocyte counts and plasma HIV load were further analyzed. Results The frequency of CXCR5+CD8+T cells increased significantly in patients with HIV infection compared with those of health controls(P<0.05).In addition, the frequency of this subset in peripheral blood was slightly positively correlated with CD4+T cell counts(r=0.349,P=0.027),and was slightly negatively correlated with plasma viral load(r=-0.377,P=0.040).The IFN-γexpression level was also negatively correlated with plasma viral load in patients with HIV infection(r=-0.514,P=0.002).While IL-10 expression level was upregulated significantly in CXCR5+CD8+T cells(P<0.05)and it was slightly negatively correlated with CD4+T cell counts(r=-0.317,P=0.046)and positively correlated with plasma viral load(r=0.670,P=0.002)in HIV infected patients. Conclusions The changes of the frequency and function of CXCR5+CD8+T cells in peripheral blood are closely correlated with the progression of HIV infection.
