摘 要:采用匙吻鲟(Polyodon spathula)脾脏细胞系PSS1,运用细胞电阻抗传感(electric cell-substrate impedance sensing,ECIS)技术和传统的细胞计数法分别探讨匙吻鲟脾脏细胞系PSS1的生长特性,并通过比较两者的生长曲线图,分析两种研究方法所获结果的一致性.结果表明,ECIS技术和细胞计数法获得的生长曲线结果具有较好的一致性,且与传统的细胞计数法相比,ECIS技术是完全自动化的分析方法,实验数据有很好的重复性,有利于进行统计分析.研究结果显示,ECIS检测技术不仅可以替代传统的细胞计数法用于细胞生长特性的研究,而且在其他基础研究和药学领域也将得到广泛应用.Counting cells with a blood cell count plate is a common method of detecting cell proliferation,plotting cell growth curves,and studying cell growth characteristics.Electric cell-substrate impedance sensing(ECIS)technology is a highly innovative electrical methods,which can quantifying cell dynamic behavior without mark and damage.To determine the agreement between the two methods,the growth curves had been compared which determined by the ECIS technology monitoring system and the traditional cell counting method respectively in cultured paddlefish(Polyodon spathula)spleen cell line PSS1.The results demonstrated that the growth curves obtained with the ECIS technology monitoring system and the traditional cell counting method were in close agreement.Compared with traditional counting assay,the ECIS technology is a completely automated analysis method,and the experimental data has good repeatability,which is conducive to statistical analysis.Research results suggested that ECIS detection technology can not only replace the traditional cell counting method to study the growth characteristics of cells,but also has a wide range of applications in basic research and pharmaceutical fields.
