摘 要:目的:采用星点设计-效应面法优化苍桂颗粒的提取工艺。方法:通过单因素试验考查浸泡时间、提取时间、加水倍数和提取次数对苍桂颗粒中木兰脂素、绿原酸、桂皮醛含量的影响。基于单因素以及体外抑菌实验的结果,应用星点设计考察提取时间、加水倍数、浸泡时间3个因素对苍桂颗粒中木兰脂素、绿原酸、桂皮醛含量以及抑菌作用的影响,对结果进行二项式方程拟合,用效应面法预测最佳水提工艺。结果:二项式拟合方程预测性良好,理论预测值与实际值偏差较小。在最佳提取工艺条件(浸泡时间为60 min,8倍量水提取2次,每次提取80 min)下,木兰脂素平均含量为9.58 mg·g^(-1),绿原酸平均含量为11.49μ·g^(-1),桂皮醛平均含量为11.33 mg·g^(-1),抑菌作用平均评分为88.11%。结论:星点设计-效应面法可以准确快速地优化苍桂颗粒的提取工艺。Objective:This paper aims to optimize the extraction process of Canggui granule by using star point design-effect surface method.Methods:the effects of immersion time,extraction time,extraction multiples and extraction times on the content of magnoliin,chlorogenic acid and cinnamic aldehyde were studied by single factor test.Based on the results of single factor and bacteriostatic test in vitro.The effects of the study on the time of extraction time,the time of addition and the time of soaking were studied.The effect of the three factors on the content of magnoliin,chlorogenic acid,cinnamic aldehyde and bacteriostasis was studied,the results were binomial equation fitting,by using the response surface method to predict the best water extraction process.Results:The predictive value of the binomial fitting equation was relatively low.In the best extraction technology conditions(soaking time for 60 minutes,eight times the amount of twice water extraction,extraction of 80 minutes every time),the mean content of magnoliin was 9.58 mg·g-1,the average content of chlorogenic acid was 11.49μ·g-1,the average content of cinnamic aldehyde was 11.33 mg·g-1,and the antibacterial effect was 88.11%.Conclusion:The design of star point design-effect surface method can optimize the extraction process of the Canggui granules accurately and rapidly.
