摘　　要：目的检测乙醛脱氢酶2(ALDH2)在肾透明细胞癌中的表达,分析ALDH2与肾透明细胞癌临床病理特征之间的关系及临床意义。方法收集17例肾透明细胞癌患者的病理标本,采用实时荧光定量PCR方法检测ALDH2在肾透明细胞癌中的表达。收集90例肾透明细胞癌患者的石蜡标本定制组织芯片,免疫组织化学法检测肾透明细胞癌组织及癌旁组织中ALDH2蛋白的表达。结果 ALDH2在肾癌组织相比癌旁组织,表达差异有统计学意义(P<0.05)。UICC分期T1期和>T1期的低表达率分别为57.38%(35/61)、63.64%(14/22),两组比较差异无统计学意义(P>0.05)。Fuhrman分级Ⅰ级和>Ⅰ级的肾癌组织中,低表达率分别为57.78%(26/45)、78.57%(33/42),差异有统计学意义(P<0.05)。ALDH2表达与肾癌患者性别、年龄、肿瘤大小及部位无统计学差异(P>0.05)。术后随访2~106个月,根据ALDH2高表达组和低表达组生存情况绘制KaplanMerier曲线及进行Log-rank检验,患者生存分析结果无统计学差异(P>0.05)。结论 ALDH2在肾透明细胞癌组织中低表达,其表达与病理分级相关。Objective To explore the expression of ALDH2 in renal cell carcinoma(RCC)and its relationship with the clinicopathological parameters.Methods The expressions of ALDH2 in 17 RCC tissues as well as matched adjacent normal tissues were detected with RT-qPCR.The protein expression of ALDH2 in 90 RCC specimens and paired normal para-carcinoma tissues were detected with immunohistochemical method.The results of RT-qPCR between RCC and matched adjacent normal tissues were compared with paired test.The relationship between ALDH2 expression and the clinicopathological parameters was analyzed with chi-square test.Results The ALDH2 expression was significantly down-regulated in RCC tissues than in paired normal tissues(P<0.05).The ALDH2 expression was 57.38%(35/61)and 63.64(14/22)respectively in UICC T stage and>T stage carcinoma;there was no significant difference(P>0.05).The ALDH2 expression was 57.78%(26/45)and 78.57%(33/42)respectively in Fuhrman grade 1 and>1 grade carcinoma;there was significant difference between the two groups(P<0.05).The ALDH2 expression was not related to patients gender,age,tumor size or location(P>0.05).According to a 2~106-month follow-up,the Kaplan-Merier curve of ALDH2 high expression group and low expression group was drawn and tested with long-rank test,and no difference was observed(P>0.05).Conclusion The expression of ALDH2 is significantly down-regulated in clear renal cell carcinoma and it is related to the pathological grade.
