摘 要:目的:建立HPLC法测定回神颗粒中葛根素的含量。方法:采用WondaSil C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-水(25∶75),检测波长为250 nm,流速:1.0 ml/min,进样量10μl。结果:葛根素在8.12~162.43μg/ml范围内线性关系良好(r=0.999 9),平均加样回收率为99.34%(RSD为1.49%)。结论:所建立的葛根素的含量测定方法简便快捷,专属性强,可有效控制回神颗粒的质量。Objective:To establish an HPLC method for the determination of puerarin in Huishen granule.Method:The content of puerarin in Huishen granule were determined by HPLC with a WondaSil C 18 column(250 mm×4.6 mm,5μm)and methanol-water(25∶75)as mobile phase at a flow rate of 1.0 ml/min.The detection wavelength was set at 250 nm;sample size was 10μl.Result:The optimal extraction conditions were as follows:Puerarin extracted from 0.5 g of Huishen granule powder with 25 ml of 30%ethanol solution,and then hot reflux extraction for 30 min.Puerarin was determined satisfactorily with an excellent linearity over the concentration range of 8.12~162.43μg/ml(r=0.999 9),and the average recovery rate was 99.34%(RSD of 1.49%).Conclusion:The established HPLC method is simple,rapid,and accurate,which could be used for quality control of Huishen granule.
