摘 要:目的比较研究《中华人民共和国药典》规定的3种基源黄精的化学成分差异。方法利用超声法提取收集到的17批黄精药材,提取液经PS/AL固相萃取小柱净化后,采用超高效液相色谱串联四极杆-静电场轨道阱高分辨质谱(UPLC-Orbitrap MS)技术分别对其化学成分进行表征,并用多元统计分析处理数据,包括主成分分析(PCA)、偏最小二乘法判别分析(PLS-DA)、正交校正的偏最小二乘分析(OPLS-DA)及聚类分析(HCA)。结果从正、负离子模式下的PCA、PLS-DA、OPLS-DA散点图及HCA树状图可见,3种法定基源黄精明显聚为3类。结论 3种基源黄精的化学成分存在明显差异,所建立的分析方法可用于区分3种法定基源黄精。本研究结果可为黄精药材的深入研究及质量标准的制定提供依据。Objective To comparatively study the chemical constituents of the three official materials of Polygonati Rhizoma recorded in the Chinese Pharmacopoeia.Methods The 17 batches of medicinal materials were extracted by ultrasonic method.The extracts were purified by PS/AL column of solid phase extraction,and then analyzed by ultra-high performance liquid chromatography tandem electrostatic field orbital trap high-resolution mass spectrometry(UPLC-Orbitrap MS)to profile their chemical constituents.After that,the obtained data were treated by using multivariate statistical analysis,including principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA),orthogonal partial least squares discriminant analysis(OPLS-DA)and hierarchical cluster analysis(HCA).Results Three official materials of Polygonati Rhizoma clearly clustered as three classes,which were showed in the PCA,PLS-DA and OPLS-DA scatter plots and HCA dendrogram under both positive and negative ion mode.Conclusion There is significant discrepancy in the chemical constituents of the three official materials of Polygonati Rhizoma,and the analytical methods can be used to distinguish the three official medicinal materials.The results will provide basis for the further investigation and scientific development of quality standards of Polygonati Rhizoma.
