摘  要:目的:探讨重度烧伤早期大鼠冠状动脉组织内BK通道的表达及意义。方法:将18只雄性成年SD大鼠随机分为假伤组(sham组)、烧伤组(burn组)、药物组(NS1619组)。采用大鼠95℃热水浴18s,制成烧伤面积占总体表面积30%的Ⅲ度烧伤模型。伤后12h分别取各组大鼠冠脉及血清,酶联免疫吸附试验法检测血清肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)含量;采用实时荧光定量RT-PCR法和蛋白免疫印迹法检测冠脉组织中BK通道α亚基mRNA和蛋白表达水平。结果:(1)burn组大鼠血清中LDH、CK-MB含量分别为(1793±54)、(2434±79)显著高于sham组(343±42)、(498±62)(P<0.01);(2)burn组大鼠冠脉组织中BK通道α亚基mRNA和蛋白质水平表达量显著低于sham组(P<0.05);(3)NS1619组大鼠血清中LDH、CK-MB含量分别为(459±68)、(813±84),较burn组(1793±54)、(2434±79)显著降低(P<0.01)。结论:BK通道在重度烧伤后可能通过调控冠状动脉的活动性,参与心肌细胞损伤的作用。Objective:To investigate the expression and effect of BK channel in coronary artery of severely burned rat.Methods:Eighteen rats were randomly divided into three groups:sham group,burn group and NS1619 group.Rats were subjected to full-thickness burn injury of a 30%total body surface area via exposure to water maintained at 95℃for 18 seconds.Rats were sacrificed 12 hours after burn and their coronary artery tissues and serums were sampled.The levels of CK-MB and LDH in serums of sham and burn rats were detected by ELISA;The mRNA and protein levels of BK channelαsubunit in the coronary artery tissues were examined by RT-PCR and Western blot.Results:①The values of CK-MB and LDH in serum of the burned rats were(1793±54)and(2434±79)respectively,much higher than(343±42),(498±62)in sham rats(P<0.01).②The mRNA and protein expressions of BK channelαsubunit in burn rats were much higher than those in sham rats(P<0.05).③The activity of CK-MB and LDH in burn rats(459±68),(813±84)pretreated with NS1619 was significantly decreased than the rats in burn+saline group(1793±54),(2434±79)(P<0.01).Conclusion:BK channel might regulate the reactivity of coronary artery and protects the myocardial from injury after severe burn.
