摘 要:目的探讨6-姜酚对丙酮醛诱导的HK-2肾小管上皮细胞(HK-2细胞)损伤的保护作用。方法采用丙酮醛诱导的HK-2细胞构建细胞损伤模型,并用6-姜酚进行干预。应用CCK-8法检测HK-2细胞的存活率;DAPI染色法检测HK-2细胞凋亡形态学变化。Western blot法检测活化的caspase-3蛋白表达。结果与对照组相比,模型组细胞存活率下降,差异有统计学意义(P<0.05),并出现明显凋亡特征;而加入6-姜酚处理后,细胞存活率及活化的caspase-3蛋白水平降低,差异有统计学意义(P<0.05)。结论 6-姜酚对丙酮醛诱导的HK-2细胞损伤具有保护作用。Objective To observe the protective effect of 6-gingerol on the injury of HK-2 cells induced by methylglyoxal.Methods HK-2 cells were incubated with 400μmol/L methylglyoxal for 24h and treated with different concentrations of 6-gingerol(1,10,50,100,200μmol/L).Cell viability was measured by CCK-8 assay.The morphological changes in apoptotic cells were evaluated by DAPI.The protein level of activated caspase-3 was tested by western blot.Results Compared with control group,cell viability of HK-2 cells were significant reduced by methylglyoxal;moreover,the apoptotic cells and protein level of activated caspase-3 was increased after incubated with methylglyoxal,which was inhibited by treatment of 6-gingerol.Conclusion 6-gingerol had a protective effect on HK-2 cells against the injuries induced by methylglyoxal through raising cell viability and inhibiting apoptosis.
