摘  要:目的探讨中性粒细胞性哮喘小鼠气道炎症和Th17、Treg特异性细胞因子IL-17、IL-10的变化及意义。方法将20只健康雌性小鼠随机分为2组:正常对照组(NC组)、中性粒细胞性哮喘模型组(NA组),每组10只。采用卵清蛋白(OVA)联合脂多糖(LPS)气道滴入致敏、OVA雾化激发方法建立NA小鼠模型。末次激发24小时后雾化吸入乙酰甲胆碱并测定气道反应性;收集支气管肺泡灌洗液(BALF),行白细胞总数及分类计数并检测细胞因子IL-17、IL-10含量;取肺组织HE染色观察病理改变。结果 (1)与NC组相比NA组气道反应性增高;白细胞总数、中性粒细胞数、嗜酸粒细胞数升高,且NEU计数高于EOS计数;BALF中IL-17浓度升高、IL-10浓度降低(P<0.05)。(2)肺组织病理示NA组支气管周围、血管周围大量炎症细胞浸润。结论 TH17/Treg细胞失衡可能为引起NA小鼠气道高反应性及气道炎症的原因。Objective To investigate the changes of airway inflammation and the changes of Th17,Treg specific cytokines IL-17 and IL-10 in mice with neutrophilic asthma.Methods Twenty male mice were randomly divided into 2 groups,including the normal control group,the neutrophil asthma model group(group NA),with 10 mice in each group.Using ovalbumin(OVA)combined with lipopolysaccharide(LPS)method to establish sensitized asthmatic mice.Twenty fourhours after the last challenge,airway responsiveness to methacholine(Mch)was measured,bronchoalveolar lavage fluid(BALF)was collected,and lung tissue was taken for HE staining.Wright’s staining was performed on BALF for white blood cell differential count.The ELISA method was used to determine the content of IL-17 and IL-10.Results①Compared with group NC,airway responsiveness increased,leukocyte count,neutrophil count,eosinophil count increased,and NEU count was higher than EOS count in NA group;IL-17 concentration increased and IL-10 concentration decreased in BALF(P<0.05)②The lung pathology showed that there was a large number of inflammatory cell infiltration around the bronchi around the NA group.Conclusion The imbalance of TH17/Treg cells may be the cause of airway hyperresponsiveness and airway inflammation in NA mice.
