摘  要:通过生物检材中乌头碱成分的快速检测,对法医毒物分析实验教学方面进行了探索和研究,在检测方法、沉淀剂、基质效应和检材新鲜度等方面进行了考察,提出检材腐败标志物1-甲基海因与乌头碱的相对含量变化关系。结果表明,采用高效液相色谱-质谱法可以实现生物检材中乌头生物碱的快速检测,采用乙腈–甲醇(5:1v/v)沉淀剂时蛋白沉降效果最好,可减少乌头生物碱水解,降低基质效应,提高物质回收率。该实验使学生能够根据目标物化学结构选择合适的样品前处理方法和仪器检测方法,使学生可以客观准确判断检材中目标物含量与检材新鲜度之间的关系,准确分析实验结果。该检测方法准确、快速、绿色,可作为法医毒物分析或药物分析实验教学中检测乌头生物碱等天然药物、毒物的实验方案。The rapid detection of Aconitum alkaloids in biological samples was explored in the teaching of experimental toxicology of forensic medicine,and the detection methods,precipitant,matrix effect and freshness of samples were investigated and put forward the relationship between the relative content of 1-methylhydantoin and aconitine.The results showed that the HPLC-MS could be used for the rapid determination of aconite alkaloids in biological samples.The best settling effect was acetonitrilemethanol(5:1v/v)for reducing the matrix effect and improving material recovery.In according to chemical structure of the target sample,students can choose the appropriate pretreatment methods and instrument detection methods.And students can accurately judge the relationship between the content of the target substance and the freshness of the sample,with the forensic toxicological analysis of scientific and reasonable explanation.The method is accurate,rapid and green,and can be used as an experimental scheme for the detection of Aconitum alkaloids and other natural drug poisons in the teaching of forensic toxicology analysis or drug analysis experiment.
