摘 要:在肉鸡养殖业中,脂肪沉积过多已成为很严重的问题。RB1基因作为细胞周期的负调控因子,调控脂肪细胞的增殖和分化过程。本研究以RB1基因为影响鸡脂肪生长发育的候选基因,在鸡全基因组SNP芯片上筛选到RB1基因上的4个SNP位点的基因型。比较RB1基因上4个SNP位点等位基因频率在高、低脂系间的差异,结果发现其中3个SNP位点(Gga_rs13552715、GGalu GA054680和GGalu GA054691)的等位基因频率在肉鸡高、低脂系间存在显著差异(P<0.05)。进一步分析SNP多态性与鸡腹脂重和腹脂率的相关性,结果发现GGalu GA054691位点多态性与腹脂重显著相关(P<0.05),Gga_rs13552715和GGalu GA054667位点多态性与腹脂重有一定程度的相关(P<0.2);GGalu GA054691和Gga_rs13552715位点多态性与腹脂率显著相关(P<0.05),GGalu GA054667位点多态性与腹脂率有一定程度的相关(P<0.2)。上述结果说明RB1基因确实是影响鸡腹脂沉积的重要基因。本研究结果为深入研究RB1基因影响鸡腹脂性状的遗传机理提供了重要依据。In broiler breeding industry,excessive fat deposition has become a very serious problem.RB1 gene is a negative regulator of cell cycle,regulating the proliferation and differentiation of adipocytes.In this study,we chose RB1 gene as candidate genes that affect the growth and development of chicken fat.The chicken 60k SNP chip was used to get the genotypes and we found 4 SNPs on the RB1 gene in the SNP chip.The allele frequencies of the four SNPs on the RB1 gene were compared between lean and fat broiler lines and the result indicated that the allele frequencies of three SNPs(including Gga_rs13552715,GGaluGA054680,and GGaluGA054691)were significantly different(P<0.05)between the two lines.Further analysis of the associations between polymorphisms of the four SNPs and abdominal fat weight and abdominal fat percentage were carried out.The results showed that GGaluGA054691 was significantly associated with abdominal fat weight(P<0.05).The association between Gga_rs13552715 and GGaluGA054667 and abdominal fat weight reached the suggested significant level(P<0.2).GGaluGA054691 and Gga_rs13552715 were significantly associated with abdominal fat percentage(P<0.05).The association between GGalu-GA054667 and abdominal fat percentage reached the suggested significant level(P<0.2).The above results indicate that the RB1 gene is an important gene that associated with abdominal fat deposiGGalution in chickens.The study of this study provides a basis for further study on the genetic mechanism of RB1 gene in chicken fat.
