摘 要:目的评价一种新型胸腰椎前路内固定系统在体外细胞毒性和肌肉植入后的生物相容性。方法体外细胞毒性试验:采用四唑盐(MTT)比色法,以L929小鼠成纤维细胞为研究对象,观察细胞形态、计算相对增殖率并分级;肌肉植入试验:以12只SD大鼠为研究对象,将材料植入脊柱旁肌肉,试验周期为26周,观察肌肉局部组织反应。结果体外细胞毒性试验:此内固定材料在浸提液浓度为100%、75%、50%、25%时,分别具有1级、1级、1级和0级细胞毒性反应。肌肉植入试验:内固定材料组术后第1周炎性细胞反应程度和囊壁形成分别为Ⅳ级和0级,第4周的分别为Ⅱ级和Ⅱ级,第12周的分别为Ⅰ级和Ⅰ级、第26周的分别为Ⅰ级和Ⅰ级。结论全内置式可膨胀型生物涂层脊柱前路内固定系统无体外细胞毒性,在肌肉植入试验中,对周围组织无刺激。Objective To evaluate the biocompatibility of a newly developed thoracolumbar anterior inter-nal fixation system.Methods The in vitro cytotoxicity test was performed as follows.According to the tetrazolium colorimetric method,we used the L929 mouse fibroblast cells as the research object.The relative growth rate was calculated and graded,and cell morphology was observed.Muscle implantation test was performed as follows.Twelve SD rats were taken as the research subjects for 26 weeks,and the local tissue reaction of muscles was observed.Results The cells in each experimental group was graded 0~1 for in vitro cytotoxocoty.After the first week,the degree of inflammatory cell reaction and the formation of the cyst wall in the experimental group were gradedⅣand 0,respectively.Moreover,gradeⅡandⅡafter fourth weeks,gradeⅠandⅠafter twelfth weeks,gradeⅠandⅠafter twenty-sixth weeks,respectively.Conclusion This internal fixation system did not have cytotoxicity in vitro,and it has no stimulation on the surrounding tissue in muscle implantation test.
