电针对脾虚大鼠胃动力及丝裂原活化蛋白激酶信号转导通路的影响

摘　　要：目的探讨电针对脾虚大鼠胃动力的调节作用及其相关机制。方法 SD大鼠皮下注射利血平造成实验性脾虚模型,实验分为正常组、模型组、针刺组、非穴组,电针"足三里"干预7 d,观察大鼠胃电参数变化,检测胃排空率及肠推进率,血清胃泌素及胃动素水平,胃窦组织缝隙连接蛋白43(CX43)、p-细胞外信号调节激酶(ERK)1/2、p-c-Jun氨基末端激酶(p-JNK)、p-p38丝裂原活化蛋白激酶(MAPK)的蛋白表达。结果与正常组比较,模型组大鼠胃运动频率及幅度显著降低,胃排空率、血清胃泌素及胃动素水平降低,胃窦组织CX43蛋白表达明显下调;与模型组、非穴组比较,电针组大鼠胃运动频率及幅度显著升高,胃排空率及血清胃泌素、胃动素水平升高,胃窦组织CX43、p-ERK1/2、p-p38 MAPK蛋白表达明显上调。结论电针"足三里"可促进脾虚大鼠胃动力,其作用机制可能与胃窦组织CX43的高表达和MAPK信号转导通路ERK1/2及p38 MAPK磷酸化有关。Objective To investigate the regulative effects of electroacupuncture on gastric motility in rats with experimental spleen deficiency;To explore its possible mechanism.Methods Rat model of experimental spleen deficiency was established by subcutaneous injection of reserpine,and were randomly divided into control group,model group,acupuncture group,and non-acupoint group.After electroacupuncture stimulation at“Zusanli”for 7 days,the changes of gastric electrical activity,the gastric emptying and intestinal propulsion rate,the serum levels of gastrin and motilin,and the protein expressions of CX43 in gastric antrum were determined.Results Compared with control group,the frequency and amplitude of gastric motility,gastric emptying rate,the serum levels of gastrin and motilin,and the expressions of CX43 decreased in the model group.Compared with model group and non-acupoint group,the frequency and amplitude of gastric motility,gastric emptying rate,the serum levels of gastrin and motilin,and the expressions of CX43,p-ERK1/2,and p-p38 MAPK increased in the electroacupuncture group.Conclusion Electroacupuncture stimulation at“Zusanli”acupoint can promote gastric motility in rats with experimental spleen deficiency,and this effect may be attributed to the high expression of CX43,as well as ERK1/2 and p38 MAPK phosphorylation in gastric antrum.
