益气复脉冻干粉与其单味药配方液HPLC指纹图谱差异性研究

摘　　要：目的研究益气复脉冻干粉与其单味药配方液的HPLC指纹图谱差异性,为进一步探索其类致敏成分提供基础。方法采用Scienhome Kromasil ODS色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.05%磷酸为流动相梯度洗脱,流速为1.0 m L/min,检测波长206 nm,柱温30℃。结果建立了注射用益气复脉冻干粉高效快速的检测分析方法,以人参皂苷Rg1为参照物峰,确定了17个共有峰。制剂与其单味药配方液图谱比较,显示在13~17 min、46 min的峰有明显差异,提示可能是寻找类致敏成分的突破口。结论本研究建立了一种稳定、快速、有效且重复性好注射用益气复脉冻干粉的质量控制方法,同时为探索该制剂的类致敏成分提供了基础。Objective To study the difference of HPLC fingerprints between Yiqi Fumai Injection and its single herb formula liquid;To provide a basis for further study of its allergenic ingredients.Methods The HPLC analysis was performed on a Scienhome Kromasil ODS column(250 mm×4.6 mm,5μm)and the mobile phase consisted of acetonitrile-0.05%phosphoric acid by stepwise gradient elution.The flow rate was 1 mL/min and the detective wavelength was set at 206 nm.The column temperature maintained at 30℃.Results A more efficient and rapid method for the detection and analysis of Yiqi Fumai Injection was established.With ginsenoside Rg1 as the reference peak,17 common peaks were identified.The spectra between Yiqi Fumai Injection and its single herb formula liquid showed significant differences in the peaks of 13–17 min and 46 min,suggesting that it may be a breakthrough in the search for allergenic ingredients.Conclusion A stable,rapid,effective and reproducible method was established for quality control of Yiqi Fumai Injection.The study can also provide a basis for exploring the allergenic ingredients of the preparation.
