摘 要:目的:通过观察黄连素对2型糖尿病大鼠血脂调节作用及对IL-6、IL-10及CRP水平影响,探讨其对2型糖尿病的作用机制。方法:80只成年雄性健康SD大鼠,随机取20只作为对照组,60只以高糖高脂饲料喂养建立2型糖尿病(T2DM)模型,造模成功后随机分为模型组及黄连素治疗组。黄连素组给予黄连素125mg/(kg·d)灌胃,模型组和正常对照组给予同等量生理盐水,共4周。4周后处死大鼠,取血测定FBG、TG、TC、CRP、IL-6、IL-10水平。给药期间记录大鼠的体重、饮食、饮水及尿量。结果:黄连素可以改善2型糖尿病大鼠的临床症状及精神状态;黄连素组FBG、TG、TC水平低于模型组,差异有统计学意义(P<0.05);模型组和黄连素组的CRP和IL-6水平高于正常组,差异有统计学意义(P<0.05);黄连素组CRP和IL-6水平低于模型组,差异有统计学意义(P<0.05);模型组和黄连素组IL-10的水平低于正常组,差异有统计学意义(P<0.05);黄连素组IL-10水平高于模型组,差异有统计学意义(P<0.05)。结论:黄连素对2型糖尿病大鼠有治疗作用,降低血糖血脂,通过调控血清炎症因子水平发挥其降糖的作用机制。Objective:By observing the effect of berberine on blood lipid regulation and IL-6,IL-10 and CRP levels in type 2 diabetic rats,the mechanism of action of berberine on type 2 diabetes was discussed.Methods:80 male adult healthy Sprague-Dawley rats were randomized to control group(n=20),and 60 were fed with high-sugar and high-fat diets to establish type 2 diabetes(T2DM)models.After successful modeling,they were randomly divided into model group and berberine treatment group.The berberine group was given berberine 125 mg/(kg·d)by gavage,and the model group and the normal control group were given the same amount of saline for a total of 4 weeks.After weeks,the rats were sacrificed,and blood was taken to determine the levels of FBG,TG,TC,CRP,IL-6,and IL-10.The weight,diet,drinking water,and urine volume of the rats were recorded during the administration period.Results:Berberine can improve the clinical symptoms and mental state in type 2 diabetic rats;the FBG,TG,and TC levels in the berberine group were lower than those in the model group(P<0.05);the levels of CRP and IL-6 in the model group and berberine group were higher than those in the normal group(P<0.05);the levels of CRP and IL-6 in berberine group were lower than those in model group(P<0.05);the levels of IL-10 in the model group and berberine group were lower than those in the normal group(P<0.05);the IL-10 level in berberine group was higher than that in the model group.Conclusions:Berberine has a therapeutic effect on type 2 diabetic rats,reduce blood glucose and blood lipids,through the regulation of serum inflammatory factors to play its role in the mechanism of hypoglycemic.
