摘 要:目的探讨葡萄糖对BeWo细胞内脂素(visfatin)表达的影响及机制。方法用不同浓度的葡萄糖作用于Be Wo细胞48 h,RT-PCR和Western-blot分别检测visfatin、PPAR-γ mRNA和蛋白表达,Western-blot检测p-JNK蛋白表达,ELISA检测visfatin分泌水平。结果与对照组相比,5.5 mmol/L和10 mmol/L组visfatin mRNA表达升高(P<0.05),30 mmol/L组明显升高(P<0.01);PPAR-γ mRNA水平仅30 mmol/L组升高(P<0.05);二者mRNA表达正相关(r=0.686,P=0.014)。与对照组相比,10 mmol/L和30 mmol/L组visfatin蛋白表达升高(P<0.05);葡萄糖对PPAR-γ及p-JNK蛋白表达无明显影响。结论高糖可促进visfatin基因和蛋白表达,PPAR-γ可能在基因水平参与葡萄糖对visfatin表达的调控。Objective To explore the effects of glucose on visfatin expression in BeWo cells and the underlying mechanism.Methods BeWo cells were cultured in mediums with glucose of various concentrations for 48 h.We measured the mRNA levels of visfatin and PPAR-γby RT-PCR.Protein levels of visfatin,PPAR-γand p-JNK protein were measured by Western-blot.Visfatin secretion level was detected by ELISA.Results Compared with those in controls,increased visfatin mRNA levels were observed in cells treated with 5.5 mmol/L and 10 mmol/L glucose(P<0.05),whereas it increased significantly in 30 mmol/L group(P<0.01);PPAR-γmRNA level increased only in 30 mmol/L group(P<0.05).Visfatin mRNA level was positively correlated with the mRNA level of PPAR-γ(r=0.686,P=0.014).Compared with control,visfatin protein expression in cells treated with 10 mmol/L and 30 mmol/L glucose increased(P<0.05).In contrast,glucose had no significant effect on the expression of PPAR-γand p-JNK proteins.Conclusion High glucose can increase visfatin gene and protein expression,while PPAR-γmay participate in the regulation of glucose on visfatin expression at the gene level.
