摘　　要：目的探讨TNF-α椎间盘注射对大鼠软骨终板β-catenin与Adamts-5蛋白表达的影响与意义。方法取SPF级12周龄SD大鼠30只,随机分成空白对照组(A组)、假手术组(B组)、TNF-α椎间盘注射组(C组),每组10只(n=10)。C组大鼠在X线透视下穿刺椎间盘并注射TNF-α25μL(10 ng/μL),B组大鼠椎间盘穿刺成功后注射生理盐水25μL,A组大鼠椎间盘不做任何干预。术后4周采用组织病理学观察软骨终板退变情况,并采用Western-Blot检测软骨终板β-catenin与Adamts-5蛋白表达。结果 HE染色显示C组大鼠椎间盘软骨终板出现明显退行性改变。Western-Blot检测显示C组大鼠椎间盘软骨终板β-catenin与Adamts-5表达均显著上调,与A组及B组相比,差异有统计学意义(P<0.05)。结论 TNF-α椎间盘注射可诱发椎间盘软骨终板退变,同时上调软骨终板β-catenin及Adamts-5表达,提示TNF-α上调β-catenin及Adamts-5表达在软骨终板退变中可能发挥了重要作用。Objective To investigate the effect and significance of intervertebral disc injection of TNF-αon the expression ofβ-catenin and Adamts-5 protein in rat cartilage endplate.Methods 30 Sprague-Dawley(SD)rats of SPF grade were randomly divided into blank control group(group A),sham operation group(group B)and TNF-αintervertebral disc injection group(group C),with 10 rats in each group.The rats in group C were given intervertebral disc puncture under X-ray and injected with TNF-αof 25μL(10 ng/μL).Rats in group B were injected with normal saline of 25μL after successful intervertebral disc puncture.Rats in group A were not given any intervention on intervertebral disc.Four weeks after operation,histopathology was used to observe the degeneration of cartilage endplate.The expression ofβ-catenin and Adamts-5 proteins in cartilage endplate was detected by Western-Blot.Results HE staining showed obvious degenerative changes of intervertebral cartilage endplate in the rats in group C.The results of Western-Blot showed that the expressions ofβ-catenin and Adamts-5 in the cartilage endplate in group C were significantly increased compared with those in group A and B,and the differences were statistically significant(P<0.05).Conclusion Intervertebral disc injection of TNF-αcan induce degeneration of intervertebral disc cartilage endplate,and at the same time,the expression ofβ-catenin and Adamts-5 in cartilage endplate is up-regulated,indicating that up-regulation ofβ-catenin and Adamts-5 expression by TNF-αmay play an important role in the degeneration of cartilage endplate.
