摘 要:目的:探讨I^-诱导的人甲状腺原代细胞的氯通道的电生理学及药理学特性。方法:干贴壁法进行原代细胞培养;免疫荧光法鉴定甲状腺原代细胞;全细胞膜片钳技术记录细胞氯通道电流;阴离子置换法研究氯通道离子选择性。结果:细胞外灌流NaI(1μmol/L)可激活甲状腺原代细胞膜氯通道开放,产生氯电流,该电流有明显外向优势,无电压及时间依赖性失活,其翻转电位接近氯离子平衡电位。该电流可被氯通道阻断剂NPPB抑制,同时该电流具有容积敏感性,能够被47%高渗溶液抑制。该通道对阴离子的通透性依次为:I^->Br^->Cl^->Gluconate^-。结论:NaI可以激活人甲状腺原代细胞容积敏感性氯通道,该通道对I^-的通透性高于Cl^-。Objective:Current study focuses on the electronic physiological and pharmacological property of iodide-induced chloride channels in primary thyroid cells.Methods:Adherence method was used for primary culture;Primary thyroid cells were identified with the detection of thyroglobulin;Whole-cell patch-clamp technique and anion substitution experiment were applied to record chloride channel currents.Results:Chloride currents could be induced by NaI(1μmol/L)in primary thyroid cells.The currents showed significant outwards rectification and lack of obvious voltage-or time-dependent depletion with the reverse potential close to the equilibrium potential of chloride.Iodide-induced chloride currents were inhibited by chloride channel blocker NPPB and 47%hypertonic solution,illustrating the property of volume-sensitivity.The anion permeability was I^->Br^->Cl^->Gluconate-in anion substitution experiment.Conclusion:Volume-sensitive chloride channels could be stimulated by NaI,showing higher anion permeability in I^-than Cl^-.
