摘  要:目的:探讨脑损伤早期血脑屏障(BBB)结构和功能的变化。方法:采用Feeney等的自由落体撞击法建立中度创伤性脑损伤模型,伤后3h、6h取大鼠大脑皮层进行电镜观察血脑屏障的形态学改变;同时经鼠尾静脉注射^(125)I-神经生长因子(^(125)I-NGF),行伤侧大鼠大脑皮层放射性计数。结果:与假伤组比较,电镜观察损伤组脑皮层血脑屏障内皮细胞胞饮小泡增多,微绒毛和指状突起形成;损伤后脑皮层放射性计数明显增多(P<0.01)。结论:大鼠脑损伤后BBB开放,机制是内皮细胞紧密连接开放、内皮细胞转运能力增强。Objective:To study the changes of structure and function Blood-Brain Barrier(BBB)in early stage of cerebral cortex injury in rats.Methods:Brain injury animal model was established according to Feeney’s method.3 hours and 6 hours after the traumatic,brain injury model in rat was established,the pathological change of BBB was observed by transmission electron microscope.The permeation of NGF through the damaged BBB was estimated by countingγ-rays after intravenous injection 125I-NGF.Results:In ipsilateral cortex,a lot of vacuoles,villus finger-like tuber was found in endothelium of capillary wall after injury,and the tight junctions were destroyed after injury.The content of 125I-NGF increased after injury(P<0.01).Conclusion:The BBB routeway in cortex at opening time is opening of tight junction,capillary rupture and enhanced transport ability of endothelium.
