摘　　要：Smad2基因是Smads蛋白家族的成员之一,Smad家族中的信号分子对绵羊的繁殖、卵泡发育有重要影响。本实验选取24只3~4岁健康空怀敖汉细毛羊,采用荧光定量PCR、蛋白免疫印迹和免疫组织化学技术,测定Smad2基因在绵羊不同发情时期(乏情期、发情间期、发情前期、发情期)的表达量并分析其在卵泡中表达规律。结果表明:Smad2基因m RNA和蛋白均在绵羊发情前期表达量最高,m RNA水平极显著高于乏情期、发情间期和发情期(P<0.01);而发情前期蛋白水平极显著高于乏情期和发情期(P<0.01),虽高于发情间期,但差异不显著(P>0.05);Smad2蛋白主要在卵巢颗粒细胞、卵泡膜细胞以及卵母细胞表达。据此,Smad2基因可能会促进敖汉细毛羊的发情启动。Smad2 belongs to the Smads proteins family.Some studies showed that the Smads family have significant impact on sheep's breeding and follicular development.A total of 24(3 to 4 years old)unpregnant Ao fine wool ewes were selected.By using the SYBR Real-time PCR,Western blotting and immunohistochemistry techniques to study Smad2 gene expression level and pattern in sheep follicles during different estrus(anestrus,diestrus,proestrus and estrus).The study showed that the expression of mRNA and protein of Smad2 gene were highly expressed in diestrus,It was observed that the Smad2 mRNA expression level was significant higher than anestrus,proestrus and estrus(P<0.01).However,the protein level of proestrus was significantly higher than anestrus and estrus(P<0.01),it also higher than diestrus,but the difference not significant(P<0.05).The protein expression of Smad2 mainly in ovarian granulosa cells,follicular cells and oocyte.Therefore,Smad2 gene may promote estrus initiation in Aohan fine wool ewes.
