异源性单、多克隆血型检测抗体对大熊猫红细胞检测的探索性研究

摘　　要：大熊猫输血治疗中的关键环节是大熊猫血液配合性实验,由于国内外尚无专业试剂,本研究使用异源性单、多克隆抗体对大熊猫血液进行定型及相容性实验。使用人类ABO血型系统定型试剂,MN血型系统血型试剂及Rh血型系统的血型试剂对大熊猫红细胞进行检测;使用人类ABO系统标准试剂细胞对大熊猫血浆进行检测;使用小鼠、人类混合血浆及亚洲黑熊血浆与其中部分熊猫进行血液相容性检测。结果表明,多种人类血型定型单克隆抗体无法检出大熊猫红细胞抗原的存在,人类血型定型试剂红细胞及人类抗血清作为大熊猫红细胞血型试剂无法避免由种间抗体引发的凝集,故不可作为大熊猫血型定型试剂;亚洲黑熊中存在可以与大熊猫红细胞抗原发生反应的红细胞血型抗体。部分亚洲黑熊可以用于大熊猫输血异种血液来源。The typing of red blood cells(RBCs)is the key point for transfusion treatment of giant panda.Due to the lack of typing reagents for giant panda,we use heterologous monoclonal/polyclonal blood typing antibody(human blood group typing reagents)to conduct blood typing test and blood compatibility test for giant panda.We tested the red blood cell of giant panda using the human ABO blood-group system,the MN blood-group system,and the Rh blood-group system.We tested the plasma of giant panda using the standard reagent cell of human ABO system,and we tested the compatibility of giant panda blood with mixture of mice and human plasma,and with Asian black bear plasma.We found that no antigen of the giant panda RBC could be detected by human blood group reagent antibodies.Human blood group reagent cells and human antisera could not be prevented from agglutination caused by interspecific antibody and cannot be used as giant panda erythrocyte blood type reagent.In the blood of Asian black bear,there is red blood cell antibody that reacts with the red cell antigen of giant panda.Some Asian black bear can serve as a heterologous blood source for transfusion treatment of giant panda.
