摘　　要：[目的]为筛选出花魔芋花粉离体培养的适宜条件。[方法]以花粉萌发率和花粉管长度为指标,研究了不同培养基配方、培养温度、培养时间、pH值对花魔芋花粉离体萌发的影响。[结果]花魔芋花粉离体萌发及花粉管伸长的最适培养基为:蔗糖10%、硼酸H3BO3100mg·L-1、Ca(NO3)2200mg·L-1、MgSO4300mg·L-1、KNO3200mg·L-1,离体培养最适培养温度为28℃,最适观察时间为4h,最佳培养pH值为6.5。[结论]通过筛选,确定了花魔芋花粉萌发和花粉管伸长的最适培养环境条件和观察时间,研究结果为魔芋人工辅助授粉工作提供一定理论依据。[Objective]In order to determine the optimal culture conditions for pollen germination,effects of culture medium,culture time,temperature and pH value were studied using in vitro assay with Amorphophalluskonjac pollens.[Methods]Pollen germination rate and pollen tube length were used as indexes to determine the idea in vitro culture condition of pollens.[Results]The results showed that the most suitable culture medium for pollen germination and tube growth was consisted of 10%sucrose,100 mg·L^-1 boric acid,200 mg·L^-1 lime nitrate,300 mg·L^-1 magnesium sulfate,and 200 mg·L^-1 potassium nitrate.The most effective culture time was 4 hours,and the best temperature for pollen germination and growth was at 28℃with pH value at 6.5.[Conclusion]The optimal culture conditions and observation time of pollen germination and pollen tube length of Amorphophalluskonjac were determined in the present study,which provided a theoretical support for the artificial pollination study of Amorphophalluskonjac in the future.
