摘 要:目的:采用运动性骨骼肌微损伤动物模型,探讨横向张力传递在针刺干预运动性骨骼肌微损伤效应中的作用。方法:120只健康雄性wistar大鼠随机分为安静组(C)、运动组(E)、运动针刺组(EA)、运动针刺阻断剂组(EAI)。C组自由饮食,其他各组运动方式采用坡度为-16°、速度为16 m/min、时间为90 min下坡跑离心运动。EA组运动后即刻,用直径0.25 mm毫针纵向斜刺两侧趾长伸肌,留针4 min。EAI组运动后即刻,大鼠尾静脉注射TRP通道阻断剂Gd Cl3,30 min后针刺。之后,分别于1 h、48 h、120 h取大鼠两侧趾长伸肌,分别用于在体状态下测量横向张力传递变化和透射电子显微镜观察骨骼肌超微结构、相邻细胞间胞外基质结构以及肌束膜胶原纤维网络分支缆细小分枝与肌内膜、肌膜的衔接位点变化。结果:1)趾长伸肌张力αi与αi'相比统计变化:C组α3低于α3'但无显著差异(P>0.05),E和EAI组1 h、48 h、120 h均具有统计学意义(P<0.05、P<0.01),EA组于1 h、48 h分别为显著性降低(P<0.05)、非常显著性降低(P<0.01),120 h组无显著性差异(P>0.05)。2)趾长伸肌各肌腱头张力θi统计变化:(1)C组θ3与θ2相比无显著性差异(P>0.05),E和EAI组1 h呈非常显著性降低(P<0.01),EAI组48 h显著降低(P<0.05);(2)C组θ4与θ2相比无统计学差异(P>0.05),E、EA、EAI组1 h均有统计学差异(P<0.01、P<0.05)。3)趾长伸肌总横向张力传递统计变化:与C组相比,E组骨骼肌总横向张力传递大幅度增加,呈先升后降再升的趋势,EA组在1 h、120 h总横向张力低于同时相E组和EAI组,48 h则高于E组和EAI组,EA组呈先升后降的趋势,EAI组呈持续上升的趋势。4)趾长伸肌超微结构变化:与C组相比,E组骨骼肌微损伤、胶原面积、PJPs数量增加,呈先升后降的趋势,EA组骨骼肌微损伤、PJPs数量均低于E组和EAI组,EA组骨骼肌胶原面积在1 h、48 h时相均低于同时相E组,但高于EAI组,120 h低于同时相E组和EAI组Objective:Acute eccentric exercise-induced skeletal muscle damage'animal models were adopted to discuss the effect of lateral force transmission in acupuncture on treatment of exercise-induced skeletal muscle damage in this study.Methods:One hundred and twenty healthy male wistar rats were randomly divided into four groups:control group(C),exercise groups(E),exercise and acupuncture groups(EA),exercise and acupuncture inhibitor groups(EAI).C group was given free access to food and water ad libitum.Except for C group,the other groups rats ran an intermittent protocol downhill(-16°incline)at 16 m·min-1 for a total of 90 min.Manual acupuncture was applied to the extensor digitorum longus(EDL)with ashi point for 4?min using a stainless steel acupuncture needle in EA groups.EAI groups were injected with GdCl3 solution in the tail vein,then EDL were stuck with acupuncture needles with ashi point after 30 minutes.Extensor digitorum longus were examined at 1 h,48 h,120 h after the downhill running,respectively.The left EDL was used to measure lateral transmission of force,and to observe ultrastructural changes,perimysial junctional plates(PJPs)changes and extracellular matrix(ECM)changes by transmission electron microscope.Results:1)The changes of total forces transmitted to the end of the EDL during series of tenotomy(αi')and myotomy(αi)after eccentric exercise and acupuncture intervention:There is no significant difference betweenα3 andα3'in control group;There is significant differences betweenα3 andα3'in E groups and EAI groups(P<0.05,P<0.01)at 1 h,48 h,120 h;There is significant differences betweenα3 andα3'in EA groups at 1 h,48 h.2)The changes of force contribution from each head(θi)after eccentric exercise and acupuncture intervention:a)There is no significant difference betweenθ2 andθ3 in control group;There is significant differences betweenθ2 andθ3 in E group and EAI group(P<0.01)at 1 h;There is significant differences betweenθ2 andθ3 in EAI group(P<0.05)at 48 h.b)There is no significant differ
