摘  要:为了解胡黄连GPPS基因及其编码蛋白的生物学特性,并推测其功能,综合运用DNAMAN、MEGA6、Blast、Blastp和TMHMM Server等软件,分析了胡黄连GPPS基因序列及其编码蛋白的理化性质、结构域等,并对部分高等植物GPPS蛋白序列进行了比对和进化树构建。结果表明,胡黄连GPPS基因包含一个1 116 bp的开放阅读框(ORF),共编码371个氨基酸,编码蛋白为亲水性蛋白,很可能定位于叶绿体上,定位系数为6;金鱼草、小花茉莉GPPS基因编码蛋白与该蛋白相似度较高,分别为74%和77%。此外,空间结构分析和序列比对结果都表明,胡黄连GPPS基因编码蛋白与类异戊二烯合酶蛋白具有较高的相似度,推测胡黄连GPPS基因为植物萜类合成途径组分之一。In order to understand the biological characteristics of the Picrorrhiza royle GPPS gene and its encoding protein and to speculate its function,this paper synthetically analyzed the sequence of the GPPS gene,the chemical and chemical properties of the encoding protein by using DNAMAN,MEGA6,Blast,Blastp and TMHMM Server Nature,domain and so on,and some higher plants GPPS protein sequence was compared and phylogenetic tree was constructed.It was found that the GPPS gene contained a 1 116 bp open reading frame(ORF),which encoded 371 amino acids and encoded a protein as a hydrophilic protein.It was probably located on the chloroplast,the positioning factor was 6,and the protein was similar to that of the Antirrhinum mujus and Jasminum sambac,74%and 77%respectively.In addition,the spatial structure analysis and sequence alignment showed that the GPPS gene encoding protein was similar to that of isoprenoid synthase protein,suggesting that the GPPS gene was one of the plant terpene synthesis pathway components.
