摘 要:目的探讨神经调节因子1(NRG1)对老年C57BL/6J小鼠的听皮层是否有保护作用。方法选择6~8周及42~44周C57BL/6J小鼠各10只,使用RTPCR的方法分别检测了听皮层上NRG1的表达。接着随机选择42~44周C57BL/6J小鼠30只分为m-NGF组、NRG1组和生理盐水组,分别采用腹腔注射鼠神经营养因子(m-NGF)、NRG1以及生理盐水,每天1次,共2个月。通过透射电镜观察各组听皮层超微结构的改变,同时通过免疫组化的方法观察听皮层NRG1的表达改变。结果 RTPCR结果显示42~44周组C57BL/6J小鼠听皮层上NRG1的相对表达量明显低于6~8周组。与生理盐水组相比,透射电镜显示NRG1组和m-NGF组对听皮层超微结构的保护较好,免疫组化显示NRG1组的听皮层NRG1的免疫反应光密度值最高,m-NGF组次之,生理盐水组最低。结论 NRG1在C57BL/6J小鼠的听皮层上的表达随着年龄的增加而减少,补充NRG1和m-NGF后对老年小鼠的听皮层有一定的保护作用。Objective To explore whether neuregulin-1(NRG1)has protective effects on the auditory cortex of aged C57BL/6J mice.Methods We used RTPCR to detect the expression of NRG1 in auditory cortex of C57BL/6J mice of different ages(6~8 weeks,42~44 weeks;10 in each group).Three groups of 42~44 weeks C57BL/6J mice were injected intraperitoneally with the mouse neurotrophic factor(m-NGF),exogenous neuregulin,or saline for two months.We observed the ultra-structures of the auditory cortices of adult mice after treatment with transmission electron microscopy.Additionally,we observed the expression of NRG1 in the auditory cortex by immunohistochemistry.Results The expression of NRG1 in the auditory cortices of C57BL/6J mice at an age of 42~44 weeks were lower than that in 6~8 weeks-old mice.The ultra-structures of the auditory cortices,including the neurons and myelin sheaths,as scanned by transmission electron microscopy,were more complete in the m-NGF and NRG1 treatment groups than those in the saline group.The expression of NRG1 in the auditory cortex after the treatment in the m-NGF and NRG1 groups were higher than that in the saline group and that in the NRG1 group was the highest.Conclusions With increasing age,NRG1 in the auditory cortices of C57BL/6J mice is gradually reduced and NRG1 has a protective effect on the auditory cortex in adult C57BL/J mice.
