摘  要:目的观察鸡骨草对非酒精性脂肪肝(NAFLD)大鼠肝组织Rho相关激酶(ROCK)、CD14表达的影响。方法 32只SD雄性大鼠为研究对象,通过长期喂养高脂肪乳建立大鼠NAFLD模型,以体质量为基本参考,采用随机数字法分为对照组、模型组、辛伐他汀组、鸡骨草水提取物组,每组8只,连续给药8周后,检测各组的高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、甘油三酯(TG)、总胆固醇(TC);采用聚合酶链式反应(PCR)扩张法检测各组的脂多糖结合蛋白(LBP)、脂多糖CD14表达水平,采用免疫组织化学方法检测各组的ROCK蛋白表达水平,并进行比较。结果模型组的TC、TG、LDL-C水平分别为(12.45±2.54)、(10.75±1.89)、(3.87±0.53)mmol/L,显著高于对照组的(3.71±0.51)、(2.56±0.63)、(1.71±0.38)mmol/L,HDL-C水平为(1.12±0.38)mmol/L,显著低于对照组的(2.68±0.70)mmol/L,差异有统计学意义(P<0.05);辛伐他汀组、鸡骨草水提取物组TC、TG、LDL-C水平显著低于模型组,HDL-C水平显著高于模型组,差异有统计学意义(P<0.05);辛伐他汀组、鸡骨草水提取物组TC、TG、LDL-C、HDL-C水平比较,差异无统计学意义(P>0.05)。模型组的LBP和CD14表达水平显著高于对照组,差异有统计学意义(P<0.05);辛伐他汀组、鸡骨草水提取物组的LBP和CD14表达水平均显著低于模型组,差异有统计学意义(P<0.05);辛伐他汀组、鸡骨草水提取物组LBP和CD14表达水平比较,差异无统计学意义(P>0.05)。模型组的ROCK-Ⅰ、ROCK-Ⅱ表达水平显著高于对照组,差异有统计学意义(P<0.05);辛伐他汀组、鸡骨草水提取物组的ROCK-Ⅰ、ROCK-Ⅱ水平显著低于模型组,差异有统计学意义(P<0.05);辛伐他汀组、鸡骨草水提取物组ROCK-Ⅰ、ROCK-Ⅱ表达水平比较,差异无统计学意义(P>0.05)。结论鸡骨草提取物可调节NAFLD大鼠ROCK、CD14表达水平,对其肝组织具有一定保护作用。Objective To observe the effect of Abrus cantoniensis Hance on expression of Rho associated kinase(ROCK)and CD14 in liver tissues of rats with non-alcoholic fatty liver disease(NAFLD).Methods A total of 32 male SD rats were selected as research objects.The NAFLD model of rats was established by longterm feeding of high-fat milk,with body quality as the basic reference.They were randomly divided into control group,model group,Simvastatin group and water extract of Abrus cantoniensis Hance group,8 rats in each group.After 8 weeks of continuous administration,high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),triglyceride(TG)and total cholesterol(TC)were detected in each group.Polymerase chain reaction(PCR)was used to detect the expression of lipopolysaccharide binding protein(LBP)and lipopolysaccharide CD14 in each group.The expression level of ROCK protein in each group was detected by immunohistochemistry.The above observation indexes were compared.Results The levels of TC,TG and LDL-C in the model group were(12.45±2.54)mmol/L,(10.75±1.89)mmol/L and(3.87±0.53)mmol/L,which were significantly higher than(3.71±0.51)mmol/L,(2.56±0.63)mmol/L and(1.71±0.38)mmol/L of the control group,the level of HDL-C was(1.12±0.38)mmol/L,which was significantly lower than(2.68±0.70)mmol/L of the control group,the differences were statistically significant(P<0.05).The levels of TC,TG and LDL-C in the Simvastatin group and the water extract of Abrus cantoniensis Hance group were significantly lower than those in the model group,and the level of HDL-C was significantly higher than that in the model group,the differences were statistically significant(P<0.05).And there were no significant differences in the levels of TC,TG,LDL-C and HDL-C in the Simvastatin group and the water extract of Abrus cantoniensis Hance group(P>0.05).The expression levels of LBP and CD14 in the model group were significantly higher than those in the control group,the differences were statistically significant(P<0.05).T
