摘 要:目的本研究旨在探讨临床红霉素耐药粪肠球菌株生物膜的形成与耐药基因分布特点和毒力基因之间的相关性。方法收集本单位收集的251株临床分离的粪肠球菌。采用微量肉汤稀释法检测红霉素的MIC值;由结晶紫染色检测菌株的生物膜。通过聚合酶链反应(PCR)检测红霉素耐药株的耐药基因(ermA、ermB和ermC)和毒力基因(esp、asal、gelE、cylA和hyl),比较红霉素耐药菌株中生物膜阳性菌株耐药基因特点和毒力基因特点。结果在粪肠球菌临床分离菌株中,红霉素敏感、中介和耐药的粪肠球菌生物膜形成的流行率为71.4%(5/7)、51.9%(27/52)和58.9%(113/192)。ermB基因与生物膜形成相关性经统计学分析无显著相关性(P>0.05)。在肠球菌表面蛋白(esp)、表面聚集蛋白(asa1)、溶血素激活因子(cylA)阳性或透明质酸酶(hyl)、明胶酶(gelE)分离物中,生物膜的形成更为常见。结论红霉素耐药粪肠球菌生物膜形成与ermB基因无显著相关性,与esp和cylA基因有显著相关性。Objective To investigate the characteristics of biofilm formation in erythromycin-resistant E.faecalis and its relationship with resistance genes and virulence factors.Methods A total of 251 E.faecalis were collected from patients.The MIC value of erythromycin was determined by broth microdilution.Biofilms were detected by crystal violet staining.Virulence genes(ESP,asal,gelE,cylA and hyl)and erythromycin resistance genes(ermA,ermB and ermC)were detected within the genomes of the microbes by polymerase chain reaction.Results The prevalence of biofilm formation among erythromycin-sensitive,intermediate and resistant E.faecalis clinical isolates were 71.4%(5/7),51.9%(27/52)and 58.9%(113/192).There was no significant correlation between ermB gene and biofilm formation(P>0.05).Biofilm formation is more common in Enterococcus surface protein(ESP),surface aggregation protein(asa1),hemolysin activating factor(cylA)positive or hyaluronidase(hyl)and gelatinase(gelE)separation.Conclusion Erythromycin resistant Enterococcus faecalis biofilm formation had no significant correlation with ermB gene,and was significantly correlated with esp and cylA genes.
