摘 要:为了建立一种高效、简单的样品前处理方法,实现对植株中磷含量的快速检测,以微波消解溶剂硝酸体积、过氧化氢体积、消解温度及保持时间为研究因素,以添加回收率为评价指标,通过L9(43)正交试验确定微波消解植株样品的最佳条件,并对方法的准确度和精密度进行了验证。结果表明:最佳消解条件为硝酸8 m L,过氧化氢体1 m L,最高消解温度160℃,保持时间10min;该条件下所得磷的回收率为94.0%~100.7%,相对标准偏差≤2.7%,说明该方法具有较高的准确度和良好的重复性,可用于植株样品中磷含量的快速检测。Microwave digestion condition of phosphorus was studied,in order to establish a highly efficient and simple method for monitoring and determination of phosphorus content in plants.Four factors such as nitric acid volume,hydrogen peroxide volume,digestion time and holding time were chosen to study,when microwave was used for sample digestion.The optimum conditions for the digestion of phosphorus in plants were determined by using L9(43)orthogonal test design with the recovery as evaluation index.Moreover,the precision and recovery of the method were verified.The optimal conditions of microwave digestion are:the nitric acid volume for 8 mL,the hydrogen peroxide volume for 1mL,the temperature at 160℃,and holding time for 10 min.In condition,the recovery was between 94.0%and 100.7%with the RSD less than 2.7%.The experiment efficiency can be improved by using orthogonal experiment design,and this method is high accuracy,environment-friendly and time-saving.In a word,it is a reliable method for determining phosphorus in plants.
