摘 要:目的:通过建立正虚邪恋型体表瘘管大鼠模型,探讨合理的模型制作方法。方法:16只SD大鼠分为正常组与模型组,采用腹腔注射环磷酰胺联合金黄色葡萄球菌和大肠杆菌混合菌液注入埋置于大鼠背部皮下的弹簧纱条的方法建立模型,41 d后对两组大鼠的一般情况、四诊指标变化、瘘管局部表现、病理学改变、脾细胞Treg与Th17细胞含量等进行比较。结果:模型组大鼠气虚、血虚、阴虚以及阳虚程度分别为(0.81±0.10)、(0.92±0.12)、(0.89±0.06)、(0.95±0.02),正常组大鼠气虚、血虚、阴虚以及阳虚程度分别为(1.01±0.08)、(1.01±0.02)、(1.01±0.06)、(1.00±0.01),两组比较有统计学意义(P<0.05或0.01);背部瘘管病理与临床瘘管表现相符;模型组脾细胞Treg细胞以及Th17细胞含量分别为(12.03±0.76)%、(1.26±0.27)%,正常组脾细胞Treg细胞以及Th17细胞含量分别为(8.30±1.06)%、(3.38±1.04)%,两组差异有统计学意义(P<0.01)。结论:采用腹腔注射环磷酰胺联合金黄色葡萄球菌和大肠杆菌混合菌液注入弹簧纱条的方法,能制备稳定的、可重复的正虚邪恋型体表瘘管大鼠模型,为临床提供可靠的中医证型瘘管动物模型。Objective To explore a method for establishing rat subcutaneous fistula model with body resistance deficiency resulting in existence of pathogenic factor.Methods Sixteen Sprague-Dawley rats were divided into normal group and model group.The method of intraperitoneal injection of cyclophosphamide combined with Staphylococcus aureus and Escherichia coli into the spring gauze buried subcutaneously in the back of rats was used to build the model.After 41 d,the general situation,four diagnostic indicators changes,fistula manifestations,pathological changes,and Treg and Th17 cells in spleen cells of rats in two groups were compared.Results The degrees of qi deficiency,blood deficiency,yin deficiency and yang deficiency in the model group were(0.81±0.10),(0.92±0.12),(0.89±0.06)and(0.95±0.02),respectively.The degrees of qi deficiency,blood deficiency,yin deficiency and yang deficiency in the normal group were(1.01±0.08),(1.01±0.02),(1.01±0.06)and(1.00±0.01),respectively.There was statistical significance(P<0.05).The pathology of fistula was consistent with the clinical fistula.The contents of Treg and Th17 cells in spleen cells of the model group were(12.03±0.76)%and(1.26±0.27)%,and those in the normal group were(8.30±1.06)%and(3.38±1.04)%.The difference between two groups was statistically significant(P<0.01).Conclusion The method of intraperitoneal injection of cyclophosphamide combined with Staphylococcus aureus and Escherichia coli into the spring gauze buried subcutaneously in the back of rats could establish an experimental rat model of subcutaneous fistula with body resistance deficiency resulting in existence of pathogenic factor.
