重组耐热短链脱氢酶的酶学性质研究

摘  要:本文研究了重组耐热短链脱氢酶催化苯甲酰甲酸乙酯生成S-扁桃酸乙酯的酶学性质。首先研究了游离酶的p H、温度适宜范围以及不同温度下的热稳定性,接着用硅胶载体吸附法固定化酶,研究了固定化酶的一些性质。结果表明,游离酶的最适p H在6.5,游离酶在70℃下的半衰期为6.6h。硅胶能在1h内对重组耐热短链脱氢酶完全吸附,且吸附牢固。反应过程中搅动与否对固定化酶的反应速率有较大影响,低强度的搅拌即可消除外扩散阻力的影响。The enzymatic properties of the recombinant thermophilic short chain dehydrogenase,which catalyzed ethyl benzoylformate to S-Ethyl mandelate,were studied.First,we characterized the free enzyme,including its optimum temperature,and operational stability under different temperatures.Then the enzyme was immobilized on silica support by adsorption.We found the range of optimum temperature of the free enzyme was 6.0~6.3,and enzyme activity wasn’t effected by temperature between 50℃and 70℃.The stability of the enzyme increased at the temperature below 50℃.Silica support can completely adsorb the enzyme in 1 hour.Low intensity of stir was enough to eliminate the mass transfer resistence of extra-diffusion and increase the reaction rate catalyzed by immobilized enzyme.
