摘 要:研究了玻璃载银抗菌材料的Ag^+溶出性质及其与大肠杆菌作用的过程、效果和抗菌原理。通过X射线衍射仪(XRD)测试发现材料为非晶态物质,激光粒度仪分析表明材料平均粒度D_(50)在10μm以下,且粒度分布范围较宽。最小抑菌浓度方法(MIC)测试表明样品的最小抑菌浓度值为0.2mg/mL,抑菌效果显著;并发现样品粒度分布在20μm以下时对抑菌性能影响不大。采用原子吸收法分析了样品的Ag^+溶出量,发现样品Ag^+溶出量随时间延长而增加,粉体粒度越小Ag^+越容易溶出。与大肠杆菌悬液混合后混合液中Ag^+浓度明显低于纯样品,可能是被细菌吸附所致。对玻璃载银抗菌材料和大肠杆菌作用混合体进行定型、负染,并用透射电镜(TEM)观测其切片前后的形貌,结果发现玻璃载银抗菌材料处理后的大肠杆菌有菌细胞溃烂融溶、菌体断裂现象;而切片样品发现了菌内空泡变性。分析认为玻璃载银抗菌材料中Ag^+溶出,进入菌体内部,攻击细胞核质,使细菌细胞破溃死亡。The antimicrobial activity and antibacterial mechanism of as-prepared Ag-doped glass antimicrobial material against E.coli was investigated.The performance of silver ions release from the Ag-doped glass was also discussed.The sample was amorphous which was tested by X-ray diffractomer.The size D 50 of the sample powder was under 10μm and the range of particle size distribution was wider.The minimal inhibitory concentration(MIC)of the sample against the selected bacteria was 0.2 mg/mL,which mean the material had good antibacterial properties.The effect on bacteriostatic could be neglected when the sample particle size distribution under 15μm.The dissolubility of Ag^+was investigated by atomic absorption spectrometry(AAS).It was found that the dissolution amount of Ag^+was increased with time,and the smaller the particle size of the powder,the easier it is to dissolve Ag.Ag^+solution was significantly lower than that of pure sample after mixing with E.coli.The mixture of Ag-doped glass antimicrobial material and E.coli was solidified and stained,and then observed by transmission electron microscopy(TEM)before and after cutting into slices.The images showed that the treated E.coli cells were damaged and collapsed.TEM images of thin sections showed that the cytoplasm of the E.coli appeared vacuolar degeneration.It was suggested that the Ag^+dissolved into the solution,and then attacked nucleus,resulting in death of the cell.
