摘  要:目的:探讨槲皮素对细菌脂多糖(LPS)诱导流产小鼠的免疫抑制机制及其保胎作用。方法:将孕鼠随机分为对照组和实验组,对照组为PBS处理组,实验组分别为LPS流产模型组、槲皮素处理组、槲皮素+LPS处理组。每组小鼠分别在末次注射后的1、3、6、12、24h脱颈椎处死,剖腹取其子宫,观察妊娠结果。免疫组织化学方法检测CD14^+巨噬细胞数量和CD14表达量,ELISA方法检测肿瘤坏死因子-α(TNF-α)和单核细胞趋化蛋白-1(MCP-1)的含量。结果:模型组小鼠流产率和胚胎吸收率均为100%,槲皮素+LPS处理组流产率降低为40%,胚胎吸收率降至36.18%。与对照组相比,槲皮素+LPS组小鼠子宫蜕膜组织CD14^+巨噬细胞数量、CD14表达量、TNF-α和MCP-1的含量均显著增加。与模型组相比,槲皮素+LPS组小鼠各时间点CD14^+巨噬细胞数量及CD14表达量、TNF-α和MCP-1的含量均显著降低。结论:槲皮素通过抑制MCP-1的生成,从而阻止巨噬细胞向子宫内膜迁移,并且下调CD14的表达,减少TNF-α的分泌,是其对抗LPS诱导流产的免疫作用机制之一。Objective:To explore the immunosuppression mechanism and anti-abortional effect of quercetin on abortive mice induced by lipopolysaccharide(LPS).Methods:The female mice were divided into control group and experimental group randomly after pregnancy.The control was PBS treatment group,while the experimental groups were LPS abortion model group,quercetin treatment group and quercetin and LPS double treatment group respectively.Next,all the mice were put to death separately at the time of 1h,3h,6h,12h and 24h after the last treatment and their uteruses were taken out to observe the pregnancy results.At last,the number change of the CD14+macrophages and the expression of CD14 were detected through immunohistochemistry,meanwhile the expression of tumor necrosis factor-α(TNF-α)and monocyte chemoattractant protein-1(MCP-1)were also tested by ELISA.Results:The abortion rate and the embryo resorbing rate were both 100%in model group and were 40%and 36.18%respectively in quercetin and LPS double treatment group.Compared with control group,the number of CD14+macrophages,the expression of CD14,the contents of TNF-αand MCP-1 in quercetin and LPS double treatment group had significant increases.Compared with model group,the number of CD14+macrophages,the expression of CD14,the content of TNF-αand MCP-1 of quercetin and LPS double treatment group were greatly decreased.Conclusion:By inhibiting the production of MCP-1,quercetin can inhibit the migration of macrophages to the endometrium,decrease the expression of CD14 and decrease the secretion of TNF-α,which may be one of its mechanisms to go against the LPS-induced abortion.
