摘 要:目的观察聚维酮碘对浮游状态及钛合金表面的金黄色葡萄球菌细菌生物膜的体外抗菌效果,为动物实验及临床研究提供参考。方法采用细菌悬液定量杀灭实验,观察聚维酮碘对浮游状态金黄色葡萄球菌的抗菌活性;在钛合金盘表面构建24 h细菌生物膜模型,通过激光共聚焦显微镜及扫描电镜观察细菌生物膜结构;采用活菌计数法,观察聚维酮碘对钛合金表面的细菌生物膜的抗菌活性。结果有效碘浓度为0.1%,0.2%,0.3%,0.4%及0.5%聚维酮碘溶液作用30 s均可全部杀灭浮游状态的金黄色葡萄球菌。经过聚维酮碘浸泡作用5 min后,随着聚维酮碘浓度升高,钛合金盘上的菌落形成单位(CFU)计数下降。0.5%聚维酮碘对细菌生物膜浸泡作用10 min后,CFU计数为0。结论聚维酮碘不仅对浮游状态金黄色葡萄球菌具有强大的抗菌活性,对钛合金表面的24 h金黄色葡萄球菌生物膜也具有抗菌活性,且抗菌活性随着聚维酮碘浓度的增加以及作用时间的延长而增强。Objective To observe the in vitro activity of iodophor against planktonic Staphylococcus aureus and the Staphylococus aureus biofilm on surface of titanium plate,and provide reference for further animal experiment and clinical research.Methods The quantitative suspension tests were carried out to observe the antibacterial activity of iodophor against Staphylococcus aureus at planktonic state.24-h old Staphylococus aureus biofilm was establishd on the surface of titanium plate and detected by the confocal laser scanning microscopy(CLSM)and scanning electron microscopy(SEM).Colony forming unit(CFU)assays were performed to quantify the viable bactertia in the biofilm after the plates were treated with indicated iodophor.Results In laboratory conditions,all planktonic Staphylococcus aureus were killed in 30 s with 0.1%,0.2%,0.3%,0.4%and 0.5%iodophor(available iodine concentration).After immersed by iodophor for 5 min,with the continuous increase of iodophor concentration,the CFU viable count on the titanium alloy disk was declining.After immersed by 0.5%iodophor for 5 min,most of the bacteria in the biofilm were killed,but not all of them.Extending the immersing time to 10 min,the CFU became zero.Conclusion The iodophor has antibacterial activity against not only the planktonic Staphylococcus aureus,but also the 24 h Staphylococus aureus biofilm in vitro.And the antibacterial activity enhanced with the increasing of iodophor concentration and effecting time.
