元通合剂的高效液相色谱指纹图谱

摘　　要：目的建立元通合剂的高效液相(HPLC)指纹图谱,为其质量控制提供参考依据。方法采用Wonda Sil C18色谱柱(4.6 mm×200 mm,5μm),流动相为乙腈-0.1%磷酸,梯度洗脱,流速1.0 m L·min-1,检测波长280 nm,柱温30℃,进样量10μL,分析时间70 min,采用国家药典委员会"中药色谱指纹图谱相似度评价系统"(2010版)软件处理HPLC图谱。结果建立了元通合剂的HPLC指纹图谱,以黄芩苷为参照物,标出10个共有峰,分离度良好,10批元通合剂指纹图谱相似度均>0.9。结论该方法操作简便,重复性好,所建立的HPLC指纹图谱专属性强,可用于元通合剂质量的综合评价。Objective To establish high performance liquid chromatography(HPLC)fingerprint and control the quality of Yuantong mixture.Methods HPLC was applied to analyze the constituents of Yuantong mixture.The chromatography conditions were:mobile phase of acetonitrile-0.1%phosphoric acid,gradient elution,flow rate 1.0 mL·min-1,detection wavelength 280 nm,column temperature 30℃,injection volume 10μL and the running time of 70 min,using the National Pharmacopoeia Commission"HPLC Chromatographic Fingerprint Similarity Evaluation System"(2010 Edition).Results The HPLC fingerprints of Yuantong mixture were established,and 10 common characteristic peaks were demonstrated.The similarity of HPLC fingerprint was 0.9 for 10 batches of samples.Conclusion The method of HPLC fingerprint was simple and reproducible,and could reflect the quality of Yuantong mixture.
