NF-κBp65蛋白表达在大鼠晶状体损伤后促进视神经再生的研究

摘  要:目的:NF-κBp65蛋白表达在大鼠晶状体损伤后促进视神经再生情况。方法:选取成年大鼠40只随机分为A组,B组,C组,D组。将A组大鼠在正常饲养7d后全部麻醉处死(4只),将B组、C组、D组大鼠分别于术后7d、14d、21d后各麻醉处死4只。对视网膜神经节细胞分布情况和NF-κBp65蛋白表达情况行相关性分析。结果:显微镜下各组大鼠视网膜标本中被荧光金标记的RGCs形态特异,边界清晰在各时间点的分布较稳定。各组的RGCs计数在各时间点相比,P>0.05,差异无统计学意义。结论:1.视神经被钳夹损伤后,随着时间延长,RGCs计数开始不断减少,NF-κBp65表达量先上升至最高点而后开始降低。2.在视神经发生损伤后,NF-κBp65蛋白很可能在晶状体损伤中的神经再生过程发挥了重要作用,起到了保护神经节细胞的机制,这为基因转录水平靶向治疗视神经病变提供了新的思路。Objective:the expression of NF-kappa Bp65 protein promotes the regeneration of optic nerve after lens injury in rats.Methods:40 adult rats were randomly divided into A group,B group,C group and D group.All rats in group A were anesthetized and killed(4 rats)after normal 7d feeding.4 rats in group B,group C and D were anesthetized and killed after 7d,14d and 21d respectively.The distribution of retinal ganglion cells and the expression of NF--kappa Bp65 protein were analyzed.Results:under microscope,the RGCs labeled by fluorescent gold in the retina of group A and group B was specific and clear,and the distribution was clear at each time point.The RGCs count of each group was not statistically significant at all time points compared with P>0.05.Conclusion:after the 1.optic nerve was injured by clamp,the RGCs count began to decrease with time,and the expression of NF-kappa Bp65 first increased to the highest point and then began to decrease.2.after the optic nerve injury,NF-kappa Bp65 protein may play an important role in the process of nerve regeneration in the lens injury,and play a mechanism to protect the ganglion cells,which provides a new idea for the gene transcription level targeting treatment of optic neuropathy.
