摘 要:本研究通过两个试验探讨谷胱甘肽(GSH)对蒙贝利亚种公牛性控精液冻后效果的影响。试验一,在冷冻稀释液中添加GSH,最终浓度分别为0(对照)、0.5、0.75和1.0mmol/L,观察对性控精液冷冻效果的影响。结果表明,0.75mmol/L组的性控精液冻后精子活率(42.2%)极显著高于对照组(39.2%)(P<0.01);0.5mmol/L组冻后活率(40.9%)与对照组相比没有显著差异(P﹥0.05);0.75mmol/L组冻后活率优于0.5mmol/L组,但二者差异不显著;1.0mmol/L组冻后活率(34.90%)极显著低于对照组和其他试验组(P<0.01),说明GSH添加量过大对性控精液冷冻效果不利。试验二,在分离前、分离后和冷冻前分别添加GSH,研究对蒙贝利亚牛性控精液冷冻效果影响。即在原精液分离前,在染色液中添加GSH,使最终浓度达0.75mmol/L,然后进行分离;在分离后平衡前精子的缓冲液中添加GSH使其最终浓度达0.75mmol/L,然后平衡;离心后冷冻前在冷冻保护液中再次添加GSH使其最终浓度达0.75mmol/L。对照组不添加GHS,然后分别观察冻精解冻效果。结果表明,试验组性控精液冻后活率(43.5%)显著高于对照组(40.5%),此外,与对照组相比,添加GSH组的精子运动状态表现更加强劲,持续力强。两个试验结果都表明,GSH对性控精液细管剂量、前进运动精子数以及性别准确率等指标没有影响。Two experiments were conducted to investigate the effect of glutathione(GSH)on the frozen sexed semen from Montbeliard bull in this study.In experiment 1,the effects of different final concentration of GSH(0 mmol/L(control),0.5mmol/L,0.75mmol/L and 1.0mmol/L)added in the extender of freezing semen on the motility rate of frozen-thawn sexed semen were studied.The results showed that the sperm motility rate in the group of GSH dosage 0.75mmol/L(42.2%)were significantly higher than that in the control group(39.2%),but the survival rate in the group of GSH dosage 0.5mmol/L(40.9%)was not significantly different from that in the control group(P>0.05).At the same time,it was indicated that the addition dose was 0.75mmol/L better than 0.5mmol/L,but the difference between them was not significant.In addition,after the addition of 1.0mmol/L GSH,the survival rate(34.90%)was significantly lower than that of the control group and other experimental groups(P<0.01),indicating that the addition of GSH higher dose had an adverse effect on the sexed semen freezing.In experiment 2,before and after sperm sorting and before freezing,GSH was added at the different time respectively to test the freezing effect of sexed semen from Monberia bull.Firstly before the original sperm sorting,GSH was added its final concentration reached 0.75mM in staining solution,then the semen began to separate,and the sexed sperms in the buffer medium after sorting were collected,GSH was added the final concentration upto 0.75mmol/L in the buffer medium,then it was balanced and centrifuged,GSH was added the final concentration to 0.75mmol/L again in the extender of freezing semen before freezing.The living rate of sexed frozen thawed semen between the experimental group and control group was compared.The results showed that the sperm motility rate of the experimental group(43.5%)was significantly higher than the control group(40.5%).Moreover,addition of the GSH,the sperm motion performance was more strong and continuous power than control group.Results fro
