摘 要:目的探讨黄连素影响人主动脉血管平滑肌细胞(HASMCs)钙化的作用及机制。方法利用糖基化终末产物(AGEs)建立HASMCs钙化模型,予以不同浓度(10、25、50μmol/L)的黄连素干预。用冯库萨、茜素红染色观察细胞钙化程度,采用细胞内钙含量试剂盒测定钙含量,Western blotting检测各组细胞骨保护素(OPG)、骨桥蛋白(OPN)及骨形成蛋白2(BMP2)、细胞外信号调节激酶(ERK)、p38蛋白表达。结果与对照组比较,AGEs组钙化程度、细胞内钙含量升高(P均<0.05),不同浓度黄连素干预HASMCs钙含量呈浓度依赖性减少(P均<0.05);AGEs组OPG、BMP2、OPN、ERK、p38蛋白表达量较对照组明显增高(P均<0.05),不同浓度黄连素呈浓度依赖性地减少HASMCs的OPG、BMP2、OPN、ERK、p38蛋白表达量(P均<0.05)。结论黄连素可抑制AGEs诱导的HASMCs钙化,其机制可能与下调ERK、p38丝裂原活化蛋白激酶信号通路导致OPG、OPN、BMP2表达减少有关。Objective To explore the effect of berberine on calcification of human aortic vascular smooth muscle cells(HASMCs)in vitro through ERK/p38 MAPK signaling pathway.Methods The advanced glycation end-products(AGEs)were applied to establish the calcified human aortic smooth muscle cell models which were then intervened with berberine of different concentrations(10,25,and 50μmol/L).Cell calcification was observed by Von Kossa staining and Alizarin red S staining methods,and calcium concentration was determined by the cell calcium content kit.The protein expression of osteoprotegerin(OPG),bone morphogentic protein 2(BMP2),osteopontin(OPN),phosphorylation-extracellular signal-regulated kinase(ERK),and p38 protein were detected by Western blotting.Results The calcium content and cell calcification in the AGEs group were higher than those of the control group(all P<0.05),and berberine-dose dependently reduced the calcium content and the calcification of HASMCs(all P<0.05).The expression of OPG,BMP2,OPN,ERK,and p38 increased significantly in the AGEs group(all P<0.05),and berberine-dose dependently down-regulated the expression of OPG,BMP2,OPN,ERK,and p38 of HASMCs(all P<0.05).Conclusion Berberine may inhibit the calcification of HASMCs induced by AGEs through down-regulating the ERK and p38 MAPK signal pathway and thus decreasing the OPG,BMP2,and OPN expression.
