摘 要:目的探讨益智仁联合乌药对高糖培养肾小球系膜细胞增殖的抑制作用及其机制。方法取益智仁10 g、乌药10 g及益智仁10 g+乌药10 g分别加10倍量水煎煮过滤制备成浓度分别为0、25、50、100、200、400μg/mL的药液。选取肾小球系膜细胞培养后分为正常对照组、高糖组、高糖+益智仁组、高糖+乌药组、高糖+益智仁+乌药组,采用以上药物进行相应干预,并检测各组细胞增殖抑制率、miRNA-21表达及磷酸酶张力蛋白同源基因(PTEN)、磷脂酰肌醇-3激酶(PI3K)、蛋白激酶B(AKT)、p-AKT蛋白表达。结果益智仁、乌药、益智仁+乌药对高糖培养的肾小球系膜细胞均有抑制作用,且抑制作用呈浓度依赖性;药物浓度相同情况下,高糖+益智仁+乌药组系膜细胞增殖抑制率最高(P均<0.05)。选择试验药物浓度为100μg/mL,正常对照组、高糖组、高糖+益智仁、高糖+乌药、高糖+益智仁+乌药组系膜细胞miRNA-21表达分别为1.004±0.109、1.723±0.227、1.533±0.279、1.611±0.230、1.196±0.149;与其他组比较,高糖+益智仁+乌药组系膜细胞miRNA-21表达最低(P均<0.05)。高糖+益智仁+乌药组系膜细胞p-AKT、AKT、PI3K蛋白表达降低及PTEN蛋白表达升高程度与高糖+益智仁组、高糖+乌药组比较差异均有统计学意义(P均<0.05)。结论益智仁联合乌药能有效抑制高糖诱导的肾小球系膜细胞增殖,其机制与降低系膜细胞miRNA-21表达,进而影响PTEN/PI3K/AKT信号途径有关。Objective To investigate the inhibitory effect and mechanism of Yizhiren combined with Obtusiloba on the proliferation of high glucose-cultured diabetic nephropathy mesangial cells.Methods Ten g Yizhiren,10 g Obtusiloba,and 10 g Yizhiren combined with 10 g Obtusiloba were added with 10 times of water respectively to prepare the drug solution with concentrations of 0,25,50,100,200 and 400 ug/mL.After the culture,the glomerular mesangial cells were divided into the normal control group,the high glucose group,the high glucose+Yizhiren group,the high glucose+Obtusiloba group,the high glucose+Yizhiren-Obtusiloba group,and the intervention of the corresponding drugs were done in the above groups.Cell proliferation inhibition ratio,miR-21 expression and PTEN,p-AKT,AKT and PI3K protein expression was detected in each group.Results Yizhiren,Obtusiloba and Yizhiren-Obtusiloba had inhibitory effects on mesangial cells cultured in high glucose,and in concentration-dependent mannner.Under the same drug concentration,the proliferation inhibition rate in the high glucose+Yizhiren-Obtusiloba group was highest(P<0.05).When we chose 100 ug/mL as the experimental drug concentration,the expression of miRNA-21 in the mesangial cells of the normal control group,high glucose group,high glucose+Yizhiren,high glucose+Obtusiloba and high glucose+Yizhiren-Obtusiloba group was 1.004±0.109,1.723±0.227,1.533±0.279,1.611±0.230,and 1.196±0.149;compared with the other groups,the expression of miRNA-21 in the mesangial cells was the lowest in the high glucose+Yizhiren-Obtusiloba group(P<0.05).The decrease of p-AKT,AKT and PI3K protein expression and the increase of PTEN protein expression in the mesangial cells from the high glucose+Yizhiren-Obtusiloba group were statistically significant different from those of high glucose+Yizhiren group and high glucose+Obtusiloba group(all P<0.05).Conclusion The combination of Yizhiren and Obtusiloba can effectively inhibit the high glucose-induced proliferation of glomerular mesangial cells,and its mec
