地塞米松温敏水凝胶的制备及其灭菌方式和细胞毒性研究

摘  要:采用溶剂挥发法制备地塞米松温敏水凝胶。考察紫外灭菌、臭氧灭菌及电子束辐照灭菌对地塞米松温敏水凝胶的灭菌效果,并对灭菌后地塞米松温敏水凝胶的细胞毒性进行了评价。结果显示,电子束辐照灭菌后水凝胶中的细菌和真菌数为0,说明电子束辐照灭菌对地塞米松温敏水凝胶灭菌效果良好。高、中、低剂量的供试品和无药组凝胶的相对增值率(RGR)分别为96.8%、87.0%、81.6%及81.2%,说明高、中、低剂量和无药组的地塞米松温敏水凝胶细胞毒性较小,其细胞毒性符合相关要求。Dexamethasone temperature-sensitive hydrogel was prepared by the solvent evaporation method.The sterilization effects of ultraviolet,ozone and electron beam irradiation sterilization methods for gel were studied,respectively.And the cytotoxicity of the gel was evaluated after sterilization.The results showed that both the numbers of bacteria and fungi in hydrogel were zero after sterilized by the method of electron beam irradiation,which indicated that electron beam irradiation sterilization was an effective sterilization method for dexamethasone temperature-sensitive hydrogel.The relative growth rate of the high,medium,low dose and no drug groups were 96.8%,87.0%,81.6%and 81.2%respectively,which indicated that the cytotoxicity of dexamethasone temperature-sensitive hydrogel was low,and the cytotoxicity of the gel meets the requirements.
