摘  要:为考察羧甲基壳聚糖阳离子浓度对止血效果的影响,制备了一系列不同质量分数(0.5%、1%、2%、4%、8%)的京尼平-羧甲基壳聚糖水凝胶。用茚三酮法测定阳离子质量分数,考察不同京尼平物质的量浓度对羧甲基水凝胶中自由氨基质量分数的影响。研究羧甲基壳聚糖的阳离子质量分数对体外凝血指数的影响,考察阳离子浓度与凝血酶原时间(prothrombin time,PT)、凝血酶时间(thrombin time,TT)、血小板的活化聚集、红细胞的关系。结果表明,随着京尼平质量分数的增加,游离氨基的质量分数逐渐降低。止血效果随着京尼平浓度的增加而逐渐增强,8%京尼平-羧甲基壳聚糖水凝胶的止血性能最佳。其止血机理为壳聚糖游离的氨基引起内源性止血途径的激活,同时引起血小板聚集活化及红细胞的激活。To examine the effect of cationicity on haemostatic activities,we prepared carboxymethyl chitosan hydrogels with different cationicity through crosslinking with different concentrations of genipin(0.5%,1%,2%,4%and 8%).The percentage of the free amino groups in the carboxymethyl chitosan hydrogels after crosslinking was evaluated by the ninhydrin assay.The effect of cationicity on the coagulant activities was systematically studied through examining its influence on blood clot index,prothrombin time(PT),thrombin time(TT),aggregation and activation of platelet,and activation of erythrocyte.The results indicate that the content of free amino groups gradually decrease with the increase genipin concentration.The hemostatic effect is gradually enhanced with the increase of genipin concentration,and the hemostatic effect of 8%genipin-carboxymethyl chitosan hydrogel is the best.The mechanism of hemostasis is the activation of endogenous hemostasis pathway by chitosan free amino groups,at the same time,the activation of platelet aggregation and activation of red blood cells can be induced.
