摘  要:为研究核苷酸结合寡聚化结构域蛋白(nucleotide-binding oligomerization domain proteins,NODs)在磨损颗粒激活巨噬细胞引起的人工关节假体无菌松动中的作用,收集髋关节周围组织共32例,其中髋关节假体无菌性松动组(A组)7例,股骨颈骨折组(B组)8例、股骨头缺血性坏死组(C组)13例、化脓性感染组(D组)4例。采用免疫组织化学染色法检测对比各组织巨噬细胞中NOD1、NOD2的表达情况;并在体外用钛颗粒刺激小鼠RAW264.7巨噬细胞检测其NOD1、NOD2及相关炎症因子的表达。结果表明:免疫组化提示,A组巨噬细胞中NOD1和NOD2的表达水平显著高于其他各组(P<0.05)。体外钛颗粒刺激巨噬细胞后,NOD2、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)的mRNA表达在1h达到高峰,明显高于对照组;而NOD1的mRNA表达无显著差异(P>0.05)。但钛颗粒刺激巨噬细胞24h后NOD1、NOD2和TNF-α蛋白表达水平增加。NOD1和NOD2可能参与了磨损颗粒激活巨噬细胞诱导人工关节无菌性松动的过程。To investigate the role of nucleotide-binding oligomerization domain proteins(NODs)in aseptic joint loosening mediated by wear particles,totally 32 cases of tissues fromhip were collected,including 7 cases of periprosthetic tissues from aseptic joint loosening(group A),8 cases from femoral neck fracture(group B),13 cases from femoral head ischemic necrosis(group C),and 4 cases from purulent infection(group D).Immunohistochemical staining was used to detect the expression of NOD1 and NOD2 in macrophages of various tissues.In vitro,titanium particles stimulated mouse RAW264.7 macrophages were used for the detection of NOD1,NOD2 and related inflammatory cytokines expression.The results of immunohistochemistry show that the expression of NOD1 and NOD2 in macrophages of group A is significantly higher than those in other groups(P<0.05).The mRNA expression of NOD2 and tumor necrosis factor-α(TNF-α)in macrophages stimulated by titanium particles in vitro reaches a peak at 1 h,which is significantly higher than that of the control group.However,NOD1 mRNA expression has no significant difference(P>0.05).But the expression of NOD1,NOD2 and TNF-αin the macrophages stimulated by titanium particles increases after 24 h.NOD1 and NOD2 may be involved in the process of aseptic joint loosening mediated by macrophages which are activated by wear debris.
