摘 要:静电纺丝技术能很好地将各种材料制备成理想的纤维结构,模拟组织中天然的细胞外基质结构。研究通过静电纺丝的方法制备了明胶的静电纺丝膜,并以不同方式与基底材料(钛合金)结合:直接将材料电纺在钛合金表面以及通过物理吸附的方式将纺丝膜贴在钛合金表面。通过对骨髓间充质干细胞(mesenchymal stem cells,MSCs)的增殖、形态及成骨分化相关蛋白的表征,研究纺丝膜纤维结构与基底材料结合方式的选择对干细胞粘附、增殖和分化的影响。研究结果表明,明胶直接电纺在钛合金上的纤维结构立体、孔隙率较大。干细胞在其表面呈现良好的铺展,且增殖水平最佳。因此可以得出结论,明胶纺丝膜-钛合金基底具有的优良纤维结构,有利于干细胞的粘附、铺展和增殖。Electrospinning technology is a useful method for preparing variety of materials into the ideal fiber structure and simulate the structure of the natural extracellular matrix in the tissue.In this study,the electrospun film of gelatin was prepared by electrospinning,and it was combined with the substrate(titanium alloy)by different ways,in which The electrospun material was directly electrospun on the surface of titanium alloy and attached to the titanium alloy surface by physical adsorption.The effects of the electrospun fiber structure and combination mode on cell adhesion,proliferation and osteogenic differentiation were studied by the characterization of the proliferation,morphology and osteogenic differentiation related proteins.The results show that the structure and the porosity of the direct electrospun gelatin on titanium alloy are larger.Stem cells show good surface spreading and the best proliferation level.Therefore,it can be concluded that the good fiber structure of the gelatin film titanium alloy substrate is conducive to the adhesion,spreading and proliferation of stem cells.
