金钱鱼线粒体基因的SNP检测分析

摘　　要：通过Hi-seq 2000高通量测序(high throughput sequencing,Hi-seq)获得金钱鱼的转录组数据,利用获得的转录组数据进行金钱鱼线粒体基因单核苷酸多态性(single-nucleotide polymorphism,SNP)的鉴定和分析。根据生物信息学的应用方法,对RNA-seq数据进行分析和筛选,得到金钱鱼线粒体基因组90个位点上的93个SNP。其中,在9个蛋白编码基因(ND1、ND2、COⅡ、ATP6、ND4L、ND4、ND5、ND6、Cytb)上均有SNP位点,而基因COⅠ、COⅢ和ATP8上未有检测到,并且蛋白编码基因上有5个SNP为非同义突变。本研究对SNP的测定和分析将为金钱鱼亲缘地理学和母系遗传进化等研究引入新的见解。In the present study,a rich supply of transcriptomic data about Scatophagus argus were obtained by Hi-seq 2000 high throughput sequencing(Hi-seq).Single-nucleotide polymorphisms(SNPs)of mitochondrial genome were analyzed on the basis of these data.93 SNPs were identified in 90 genetic loci throughout the whole mitochondrial genome.SNPs were detected in 9 protein-coding genes(ND1,ND2,COⅡ,ATP6,ND4L,ND4,ND5,ND6 and Cytb),and not found in the other mitochondrial genes(COⅠ,COⅢand ATP8).Nonsynonymous mutation was detected in 5 SNPs.The determination and analysis of SNP in this study will introduce new insights into the research on genetic geography and maternal genetic evolution of Scatophagus argus.
