摘  要:目的探讨吸烟者外周血单个核细胞(PBMC)差异表达的miRNA与mRNA调控网络及相关信号通路。方法收集20名健康非吸烟者与17名健康吸烟者的外周静脉血,分离PBMC后提取总RNA,分别进行miRNA和mRNA表达谱芯片检测。在差异表达的miRNA和mRNA范围内通过数据库预测miRNA对mRNA的调控关系,并进行信号通路富集分析。结果与健康非吸烟者相比,吸烟者PBMC有miRNA与mRNA的差异表达,其差异表达基因的GO富集分析表明,血小板来源生长因子信号通路、胆囊收缩素受体信号通路、T细胞活化和表皮生长因子受体信号通路是主要的信号通路。结论吸烟者PBMC中差异表达的miRNA与mRNA调控网络可能与某些疾病的潜在危险因素相关。Objective To explore the regulatory network of dysregulated microRNAs and mRNAs and related signaling pathways in peripheral mononuclear cells(PBMCs)of cigarette smokers.Methods The peripheral venous blood was taken from 20 healthy non-smokers and 17 healthy smokers.The miRNA and mRNA expression profile of PBMCs was examined by miRNA and mRNA array.The regulatory relationship between dysregulated miRNAs and mRNAs was predicted and the signaling pathways were enriched.Results Compared with the healthy non-smokers,the smokers had dysregulated miRNAs and mRNAs.The GO enrichment analysis indicated that PDGF signaling pathway,CCKR signaling map,T cell activation and EGF receptor signaling pathway were enriched in the smokers.Conclusion The regulatory network of dysregulated miRNAs and mRNAs in PBMCs of smokers may be related to some potential risk factors for certain diseases.
