摘 要:【目的】探究大鲵(Andrias davidianus)抗菌肽家族基因对嗜水气单胞菌(Aeromonas hydrophila)侵染的响应,为揭示抗菌肽家族基因的功能提供理论依据。【方法】通过腹腔注射嗜水气单胞菌感染大鲵后,采用Trizol提取组织总RNA,反转录合成cDNA,再以实时荧光定量PCR(qPCR)分析抗菌肽家族3个基因(KK1、KK5和KK8)在大鲵皮肤和肝脏器官的表达响应情况。【结果】经腹腔注射嗜水气单胞菌后,抗菌肽基因KK1、KK5和KK8在大鲵皮肤组织中的相对表达量在攻毒后12~36 h均显著高于注射PBS的对照组(P<0.05,下同);而在肝脏组织中,抗菌肽基因KK1、KK5和KK8的相对表达量分别在攻毒后24、12和24 h时显著高于对照组,其他时间点均低于对照组,说明这3个抗菌肽基因在早期积极响应嗜水气单胞菌的侵染,但相对于皮肤组织,其高表达量持续时间较短。【结论】抗菌肽家族基因KK1/KK5/KK8在嗜水气单胞菌侵染大鲵时积极作出应答,尤其在皮肤组织中的特异表达对启动机体天然免疫应答抵抗病原微生物感染具有重要作用。【Objective】The response of antimicrobial peptide family gene of Andrias davidianus to Aeromonas hydrophila was explore to provide theoretical basis for revealing the function of the antimicrobial peptides family genes.【Method】Through intraperitoneal injection of A.hydrophila,A.davidianus was infected.The total RNA was extracted by Trizol method.The RNA was turned into cDNA by reverse transcription.Real-time fluorescence quantitative PCR(qPCR)analysis of the expression response of three antimicrobial peptide family genes(KK1,KK5 and KK8)in the skin and liver of A.davidianus was conducted.【Result】After intraperitoneal injection of A.hydrophila,the relative expression of antimicrobial peptide genes KK1,KK5 and KK8 in the skin was significantly higher than that in the control group at 12-36 h after injection(P<0.05,the same below).However,the relative expressions of KK1,KK5 and KK8 in liver tissue were significantly higher than that in control group at 24,12,24 h after infection respectively,and at other time points,they were lower than that in control group.These three antimicrobial peptide genes responded positively to the infection of A.hydrophila at the early stage,but their high expression duration was shorter than skin tissue.【Conclusion】Antimicrobial peptide family genes KK1/KK5/KK8 play an important role in the immune system when A.hydrophila infects A.davidianus,the specific expression in skin plays an important role in initiating the innate immune response of the body against pathogenic microorganism infection.
