叉尾斗鱼肉瘤病的病理学检测和病原鉴定

摘　　要：为确诊叉尾斗鱼肉瘤病,采集自然患病鱼进行病理学观察,基于淋巴囊肿病毒主要衣壳蛋白基因采用PCR方法对肉瘤状增生物进行检测和进化树分析。结果显示,患病叉尾斗鱼肉瘤状增生物是由大量囊肿细胞聚集在皮肤和鳍条上形成,鳃、肝、肾、脾、肠等内脏组织器官均未观察到囊肿细胞。囊肿细胞大小为正常细胞大小的57.9~1823.8倍,细胞核不规则,细胞质内可见嗜碱性包涵体;透射电镜观察可见囊肿细胞胞质内存在大量直径约171.5nm的病毒粒子。病理观察结果符合鱼类淋巴囊肿病毒感染的临床特征,PCR方法检测到1272bp的目的片段,测序结果显示,该基因与GenBank中的淋巴囊肿病毒KJ408271.1序列相似性达99%。试验结果表明,叉尾斗鱼肉瘤病为淋巴囊肿病毒感染所致,皮肤和鳍是病毒感染的靶器官。本研究为叉尾斗鱼肉瘤病临床诊断和预防提供了试验参考。The histopathology was observed in sarcosis of paradise fish Macropodus opercularis diseased naturally by uranyl acetate and lead citrate double staining transmission electron microscope observation,and then the major caspule protein of Lymphocystis disease virus(LCDV)was amplified and analyzed by the template of lymphoid cyst cells to detect the pathogenicity of sarcosis.It was found that the sarcomalike growths were formed via the accumulation of many l3rmphoid cyst cells on skin and fin,without lymphoid cyst cells on gill,liver,kidney,spleen,and intestine.The lymphoid cyst cells had as large as 57.9-1823.8 times in area as normal cells,with irregular nucleus and basophilic cytoplasmic inclusions.The transmission electron microscope revealed that cytoplasmic virus particles were about 171.5 nm in diameter in infected cells.The target fragment of 1272 bp was 9 9%identitiy with LCDV(GenBank K J40827.1),indicating that the sarcosis of paradise fish was caused by LCDV,the skin and fins being the target organs of viral infection.
