血管紧张素Ⅱ体外诱导大鼠骨髓间充质干细胞心肌样细胞分化研究

摘　　要：目的:观察血管紧张素Ⅱ(AngⅡ)在体外将大鼠骨髓间充质干细胞(BMMSCs)诱导为心肌样细胞的作用。方法:将第3代培养的BMMSCs分为AngⅡ组(终浓度为0.1μmol/L)和空白对照组进行诱导,两组诱导24h之后吸弃诱导液,用完全培养液继续培养4周。分别用倒置相差显微镜、MTT法、流式细胞仪、免疫荧光染色法、透射电镜检测细胞的形态学变化、增殖能力、分化率、心肌肌钙蛋白I(cTnI)的表达以及超微结构。结果:BMMSCs原代培养时呈现多种形态,经过传代体积增大,AngⅡ诱导后呈长梭形且均匀一致生长。MTT法显示AngⅡ组细胞的增殖能力高于对照组。流式细胞检测示:AngⅡ组诱导分化率为(25.3±2.2)%。免疫荧光染色显示AngⅡ诱导后的BMMSCs中cTnI呈阳性表达。透射电镜观察到Z线样物质和肌丝。结论:血管紧张素Ⅱ能够体外使大鼠BMMSCs诱导分化为心肌样细胞。Objective:To investigate the effect of inducing BMMSCs by angiotensinⅡinto the cardiomyocyte-like cells in vitro.Methods:The third generation of BMMSCs were divided into AngⅡgroup(final concentration:0.1μmol/L)and control group.After induction of 24h,the induced solution of two groups were discarded and cells were cultured in 4 weeks using complete culture medium.The morphological changes,proliferation ability,cell differentiation rate,ultrastructure and expression of cardiac troponin I were detected respectively by inverted phase contrast microscope,MTT assay,flow cytometry,transmission electron microscopy and immunofluorescence staining.Results:In primary culture BMMSCs had diversity of morphology.The volume of passaged cells gradually increased.After induction,the cells became long spindle and the growth was consistent.MTT assay showed higher proliferation ability of AngⅡgroupthan that of the control group.In AngⅡgroup the differentiation ratio was(25.3±2.2)%,showing by flow cytometer.cTnI staining was positive in induced BMMSCs.Z line-like substances and myofilaments were found by transmission electron microscopy.Conclusions:AngiotensinⅡsuccessfully make BMMSCs induction into the cardiomyocyte-like cells in vitro.
