摘  要:目的探讨唐山地区汉族人群特应性皮炎(AD)发病风险与外周血IL-10基因多态性的相关性。方法选择唐山地区汉族AD患者300例作为观察组,同期体检健康者300例作为对照组。采用Sequenom Mass ARRAY SNP基因分型技术检测两组外周血IL-10基因5个SNP位点(rs1554286、rs1518111、rs3021094、rs3024490、rs1800871)基因型,分析IL-10基因多态性与AD发病风险的相关性。结果 IL-10基因5个SNP位点均符合哈德-温伯格平衡(P-HWE>0.05)。观察组IL-10基因rs1554286位点AG、GG基因型,rs1518111位点CT、TT基因型,rs3024490位点AC、CC基因型,rs1800871位点AG、GG基因型占比均高于对照组(P均<0.01);rs3021094位点GT、GG基因型占比均低于对照组(P均<0.01)。观察组IL-10基因rs1554286位点等位基因G、rs1518111位点等位基因C、rs3024490位点等位基因C、rs1800871位点等位基因G频率均高于对照组,rs3021094位点G等位基因频率低于对照组(P均<0.01)。显性、隐性、加性遗传模型下IL-10基因rs1554286、rs1518111、rs3024490、rs1800871位点均与AD发病风险增加有关(P均<0.05);rs3021094位点均与AD发病风险降低有关(P均<0.05)。IL-10基因rs1554286、rs1518111、rs3021094、rs3024490、rs1800871这5个位点具有强连锁关系。ATGAA单体型与AD发病风险增加有关,GTTAA单体型与AD发病风险降低有关(P均<0.01)。结论唐山地区汉族人群AD发病风险可能与其外周血IL-10基因rs1554286、rs1518111、rs3021094、rs3024490、rs1800871位点多态性有关。Objective To investigate the association between the risk of atopic dermatitis(AD)and the polymorphism of IL-10 gene in peripheral blood of Chinese Han population in Tangshan.Methods A toal of 300 patients with AD in Han nationality were selected as the observation group,and 300 healthy participants were collected as the control group in the same period.The genotypes of 5 SNPs(rs1554286,rs1518111,rs3021094,rs3024490,and rs1800871)of IL-10 gene in the peripheral blood were detected by Sequenom MassARRAY SNP genotyping technique.The association between IL-10 gene polymorphism and the risk of AD was analyzed.Results The five SNPs of IL-10 gene were all consistent with Hardy-Weinberg Equilibrium(P-HWE>0.05).The AG and GG genotypes of rs1554286,the CT and TT genotypes of rs1518111,the AC and CC genotypes of rs3024490,and the AG and GG genotypes of rs1800871 had higher ratios in the observation group than in the control group(all P<0.01);while the GT and GG genotypes of rs3021094 had lower ratios than the control group(all P<0.01).The frequencies of rs1554286-G allele,rs1518111-C allele,rs3024490-C allele and rs1800871-G allele in the observation group were higher than those of the control group,while the frequency of rs3021094-G allele was lower than control group(all P<0.01).Under the dominant,recessive and additive genetic models,the IL-10 gene rs1554286,rs1518111,rs3024490,and rs1800871 were all associated with an increased risk of AD(all P<0.05);while the rs3021094 was associated with a decreased risk of AD(P<0.05).The rs1554286,rs1518111,rs3021094,rs3024490,and rs1800871 of IL-10 gene had strong linkage relationships.The ATGAA haplotype was associated with an increased risk of AD,and the GTATA haplotype was associated with a decreased risk of AD(P<0.01).Conclusion The risk of AD in Chinese Han population may be related to the polymorphisms of rs1554286,rs1518111,rs3021094,rs3024490 and rs1800871 of IL-10 genes in the peripheral blood.
