摘  要:目的制备复方一枝蒿提取物固体脂质纳米粒。方法高剪切乳化超声法制备纳米粒。在单因素试验基础上,以脂质、大豆卵磷脂、吐温-80用量为影响因素,粒径、包封率为评价指标,星点设计-效应面法优化制备工艺。结果最佳处方为脂质用量154.74 mg,大豆卵磷脂用量75.18 mg,吐温-80用量6.25%,所得纳米粒为球形或类球形,大小分布均匀,平均粒径(106.70±0.28)nm,PDI 0.192±0.48,Zeta电位(-18.90±0.57)m V,包封率(90.20±0.95)%。在4℃下保存60 d、常温下保存30 d时,纳米粒粒径、包封率无明显变化。结论该方法稳定可靠,可用于制备体外稳定性良好的复方一枝蒿提取物固体脂质纳米粒。AIM To prepare Compound Yizhihao extract-loaded solid lipid nanoparticles.METHODS For the nanoparticles prepared by high-shear emulsion-ultrasonic method,consumption of lipid,soybean lecithin,Tween-80 were taken as influencing factors,together with particle size and encapsulation efficiency as evaluation indices,central composite design-response surface method was applied to optimizing the preparation on the basis of single factor test.RESULTS The optimal formulation was determined to be 154.74 mg for lipid consumption,75.18 mg for soybean lecithin consumption,6.25%for Tween-80 consumption,the obtained spherical or spherical-like nanoparticles demonstrated uniform size and distribution,with the average particle size of(106.70±0.28)nm,PDI of 0.192±0.48,and Zeta potential of(-18.90±0.57)mV.When stored at 4℃for 60 d or room temperature for 30 d,the nanoparticles’particle size and encapsulation efficiency exhibited no obvious changes.CONCLUSION This stable and reliable method can be used for the preparation of Compound Yizhihao extractloaded solid lipid nanoparticles with good in vitro stability.
