摘  要:目的:通过建立BALB/c小鼠全子宫切除实验动物模型,考察和分析该术式对小鼠TDAR的影响。方法:平行对照组和手术组经腹行全子宫切除术,非手术组不予手术;非手术组和手术组腹腔注射KLH进行免疫;流式细胞技术分析B淋巴细胞亚群变化;ELISA法检测血清雌二醇和KLH Ig G抗体含量;HE染色观察卵巢组织学改变。结果:KLH能够刺激B淋巴细胞分化增殖,手术组和非手术组较平行对照组明显升高(P<0. 05);全子宫切除术能够影响KLH Ig G抗体生成,手术组较非手术组低(P<0. 05);全子宫切除小鼠雌二醇分泌明显低于非手术组(P<0. 05);全子宫切除小鼠卵巢组织出现体积缩小,成熟卵泡形态不规则,颗粒细胞松散、凋亡等。结论:全子宫切除术通过降低卵巢雌二醇分泌水平,对小鼠TDAR实验产生抑制效应。Objective:To investigate and analyze the influence of total hysterectomy model in BALB/c mice on TDAR.Methods:Mice were randomized to parallel control,operated and no-operated group.Parallel control and operated group underwent hysterectomy.The operated and no-operated group were immunized by intraperitoneal injection of KLH;The changes of Blymphocyte subsets were analyzed by flow cytometry.The concentration of estradiol and KLH IgG antibody in serum and the histological structure of the ovary were measured.Results:KLH could stimulate the proliferation and differentiation of B lymphocyte,and the operation group and no-operated group were significantly higher than those in the parallel control group(P<0.05).Total hysterectomy could affect the formation of KLH IgG antibody,and the operation group was lower than that of the no-operated group(P<0.05).The content of estradiol in the total hysterectomy mice was significantly lower than that in the no-operated group(P<0.05).Observation on ovarian histological structure of no-operated group was normal,however it were different for parallel control and operated group,such as the loss ovarian volume,the diseases of number of mature follicle,the loosing of granule cells,and apoptosis.Conclusion:Total hysterectomy has a inhibitory effect on TDAR test in mice by down-regulate the level of ovarian estradiol secretion.
